不同訓(xùn)練強(qiáng)度影響骨腱結(jié)合部損傷的機(jī)理研究

【摘要】:研究目的:骨腱結(jié)合部損傷是一種困擾運(yùn)動訓(xùn)練和人們?nèi)粘I畹某R姄p傷,過度負(fù)荷是主要原因,其病理變化過程還不十分清楚;|(zhì)是肌腱載荷的主要承擔(dān)者,在損傷的修復(fù)中有重要的作用。本文以不同強(qiáng)度的循環(huán)載荷作為預(yù)手段,對骨腱結(jié)合部損傷的機(jī)理進(jìn)行研究探討。揭示骨腱結(jié)合部損傷發(fā)生發(fā)展過程中的病理變化,以及不同載荷量引起的各種指標(biāo)相應(yīng)的變化規(guī)律,確定干引起損傷的載荷量。 研究方法:48只新西蘭大白兔隨機(jī)分為6組,每組8只。其中一組為對照組,其余5組為實(shí)驗(yàn)組。采用循環(huán)載荷的刺激方式,強(qiáng)直收縮力的30%為100%的刺激負(fù)荷強(qiáng)度組,其它各組負(fù)荷強(qiáng)度依次采取此刺激強(qiáng)度的20%、40%、60%、80%,進(jìn)行循環(huán)載荷刺激2小時(shí)/天,3天/周。4周后宰殺取材進(jìn)行相應(yīng)的¨E染色和免疫組織化學(xué)染色。 研究結(jié)果:1.各負(fù)荷組骨腱結(jié)合部組織形態(tài)發(fā)生了改變。高運(yùn)動負(fù)荷組,肌腱細(xì)胞密度發(fā)生顯著性增加。纖維軟骨帶厚度無顯著性差異。2.Ⅲ型膠原纖維發(fā)生顯著性增加,從60%載荷強(qiáng)度開始與CON組、L20、L40載荷組出現(xiàn)顯著性差異。Aggrecan免疫組化結(jié)果顯示無顯著差異。Tenascin-C表達(dá)出現(xiàn)顯著性改變,60%載荷刺激強(qiáng)度時(shí)表達(dá)下降,L80、L100組比CON組、L20、L60載荷組顯著性升高。TGF隨負(fù)荷刺激強(qiáng)度的增加而增加,L20、L40載荷刺激強(qiáng)度與CON組比較出現(xiàn)顯著性差異,L60、L80、L100載荷刺激強(qiáng)度組與CON組出現(xiàn)非常顯著性差異。3.各指標(biāo)相關(guān)結(jié)果顯示,細(xì)胞密度與Ⅲ型膠原纖維、TGF具有非常顯著性相關(guān)。TGF與型膠原纖維和TN有顯著性相關(guān)。 研究結(jié)論:1不同載荷強(qiáng)度作用于骨腱結(jié)合部4周,L60組、L80組、L100組出現(xiàn)損傷病理變化,說明這種低強(qiáng)度循環(huán)載荷叮以引起骨腱結(jié)合部損傷的發(fā)生2運(yùn)動強(qiáng)度的不同導(dǎo)致骨腱結(jié)合部組織發(fā)生了相應(yīng)的結(jié)構(gòu)和成分改變。說明了骨腱結(jié)合部損傷程度與運(yùn)動強(qiáng)度有關(guān)。3在損傷的病理變化中,Ⅲ型膠原纖維、Tenasin-C發(fā)生了顯著性增加,說明骨腱結(jié)合部損傷的發(fā)生中,細(xì)胞外基質(zhì)具有重要作用。4不同載荷強(qiáng)度作用于骨腱結(jié)合部4周,TGF表達(dá)發(fā)生了顯著性增加,并且與細(xì)胞密度、Ⅲ型膠原纖維、TNC有顯著性相關(guān),說明TGF在骨腱結(jié)合部損傷修復(fù)過程中是重要的調(diào)節(jié)因素。
[Abstract]:Objective: bone tendon joint injury is a common injury that puzzles sports training and people's daily life. Overloading is the main cause and the pathological process is not very clear. The matrix is the main load bearer of tendon. In this paper, the mechanism of bone tendon joint injury is studied by using different cyclic load as preprocessing method, and the pathological changes during the development of bone tendon joint injury are revealed. According to the change law of various indexes caused by different load quantity, the load quantity caused by dry damage is determined. Methods Forty-eight New Zealand white rabbits were randomly divided into 6 groups with 8 rabbits in each group. One group was the control group and the other five groups were the experimental group. The other groups took 20% of the stimulus intensity in turn. The rats were subjected to cyclic load stimulation for 2 hours / day / 3 days / week for 4 weeks and then slaughtered for E staining and immunohistochemical staining. Results 1. The tissue morphology of bone tendon junctions in each load group was changed. In the high exercise load group, the density of tendon cells increased significantly, and the thickness of fibrous chondrocytes showed no significant difference .2. type 鈪,
