煤焦瀝青職業(yè)暴露人群DNA甲基化和端粒損傷研究
[Abstract]:Objective to investigate the promoter methylation and telomere damage of p16 FHIT and RASSF1A gene in occupational exposure population of coal tar pitch, and to explore the effect markers of occupational exposure of coal tar pitch. Object and method 1. Participants: one hundred and eighty cases of occupational contact with coal tar pitch were collected from 145 workers of a carbon factory in the central part of the university. All of them were examined in the physical examination department of the first affiliated hospital of our school. After informed consent, the subjects were collected peripheral blood, professional investigators to collect basic information. 2. 2. Methods: the relative telomere length of peripheral blood DNA was analyzed by real-time fluorescence quantitative PCR. The methylation level of p16 RASSF1A and FHIT gene was analyzed by real-time fluorescence quantitative methylation specific PCR. 3. Statistical analysis: according to the distribution type of quantitative data, the representation method and statistical test method were selected. The qualitative data were compared by 蠂 2 test. The test level is 偽 0. 05. Result 1. The results showed that telomere length was 0.83 (0.71-0.90) in occupational exposed population and 1.13 (0.72-1.45) in control group. The promoter methylation rate of p16 gene was 0.56 (0.13-4.95) and 0.37 (0.06-4.23) in occupational exposure group and control group, respectively, and there was no significant difference between the two groups (ZP0.163), and the methylation rate of FHIT promoter in occupational exposure group and control group was 3.22 (2.09-4.64) and 2.95 (1.45-5.88), respectively. The methylation rates of RASSF1A gene promoter in occupational exposure group and control group were 14.76 (7.26-39.09) and 19.34 (3.82-50.95), respectively. Conclusion Occupational exposure to coal tar pitch can shorten the length of DNA telomere in human peripheral blood leukocytes, while occupational exposure of coal tar pitch does not change the methylation rate of p16 RASSF1A and FHIT gene promoter.
