苦蕎醇溶粗提物對淀粉酶的影響及體內(nèi)抗氧化性研究

【摘要】:苦蕎富含以蘆丁為主的黃酮類物質(zhì),與其他谷物相比有更高的食用價值和藥用價值。由于快消化淀粉會產(chǎn)生較高的餐后血糖反應(yīng)而導(dǎo)致氧化應(yīng)激,對機體造成氧化損傷,因此本文首先以淀粉的消化率為切入點,探究了苦蕎醇溶粗提物對α-淀粉酶活性的影響,進一步采用動物實驗研究了苦蕎醇溶粗提物通過其抗氧化活性對酒精性肝損傷的保護作用。采用70%的乙醇,料液比1:35(g/m L),在80℃下提取苦蕎粉得到苦蕎醇溶粗提物及提取后的苦蕎殘粉,粗提物得率為6.26%。通過薄層層析和超高效液相色譜串聯(lián)四級飛行時間質(zhì)譜法(UPLC-Q-TOF/MS)分析苦蕎醇溶粗提物成分,可知其主要成分蘆丁和槲皮素的含量分別為:198.89 mg/g粗提物,3.56 mg/g粗提物,此外還含有其他黃酮類物質(zhì)如山柰酚-3-O-蕓香糖苷、色素、大分子糖類和酚酸類物質(zhì)等。通過對比苦蕎粉和苦蕎殘粉的雙酶水解消化曲線可知苦蕎醇溶粗提物會顯著減緩苦蕎粉的消化。進一步探究影響淀粉代謝的原因,以抑制率、抑制類型、熒光猝滅效應(yīng)實驗來分析苦蕎醇溶粗提物對α-淀粉酶的抑制作用,結(jié)果表明苦蕎醇溶粗提物對α-淀粉酶的抑制作用與其濃度相關(guān),IC50值為3.35 mg/m L,抑制類型為非競爭性抑制,并可使α-淀粉酶的熒光效應(yīng)發(fā)生靜態(tài)猝滅。小鼠的餐后血糖曲線也表明了苦蕎醇溶粗提物可顯著降低餐后血糖反應(yīng),影響了普通玉米淀粉在小鼠體內(nèi)的消化吸收。以亞鐵還原能力法(FRAP)確定了苦蕎醇溶粗提物的抗氧化性后進行動物實驗,采用高脂膳食和急性酒精飼喂C57BL/6雄性小鼠,探究苦蕎醇溶粗提物的體內(nèi)抗氧化作用。結(jié)果表明高脂飼料會顯著促進小鼠增重,苦蕎醇溶粗提物對小鼠脂代謝無顯著影響;酒精處理(最后1周)后小鼠體重減輕,而苦蕎醇溶粗提物能夠降低體重減輕的幅度,表明對酒精損傷有一定保護作用。進而分析小鼠血清中丙氨酸轉(zhuǎn)氨酶(ALT)和天冬氨酸轉(zhuǎn)氨酶(AST)的活性,酒精處理后,他們的活性顯著上升,說明肝功能受損,但飼料中的苦蕎醇溶粗提物能有效抵抗ALT和AST水平的升高,也表明了苦蕎醇溶粗提物對酒精肝損傷的保護作用。通過分析肝臟和小腸組織中的超氧化物歧化酶(SOD)活力、谷胱甘肽過氧化物酶(GSH-Px)活力與丙二醛(MDA)含量,結(jié)果表明高脂膳食和急性酒精處理會造成小鼠肝臟和小腸抗氧化能力的下降,出現(xiàn)氧化損傷,而苦蕎醇溶粗提物有效提高了組織的抗氧化效應(yīng),小腸中的抗氧化性尤其顯著。通過對比小鼠的肝臟和腹部脂肪組織切片可知,酒精處理會造成小鼠的肝臟受損,苦蕎醇溶粗提物可緩和肝細胞間的炎性浸潤現(xiàn)象,表明對肝臟有一定的保護作用,而粗提物的抗氧化活性可能是重要的機制之一,值得進一步探究。
