孕鼠暴露雙酚A對仔鼠存活率、生殖激素及相關(guān)基因的影響

【摘要】:為了研究孕鼠在孕期暴露雙酚A(bisphenol A,BPA)對仔鼠生殖激素及相關(guān)基因的影響,試驗將40只昆明孕鼠隨機分為A、B、C、D共4組,每組10只。其中A組為對照組,飼喂普通鼠糧;B、C、D組孕鼠整個妊娠期(妊娠1d至分娩)分別按每只鼠每天50、500、2 500mg/kg體重給予BPA,待孕鼠自然分娩,觀察記錄仔鼠死亡情況。至仔鼠性成熟(56日齡)剖殺仔鼠,摘取睪丸或卵巢稱重并計算臟器指數(shù),HE染色觀察卵巢或睪丸組織結(jié)構(gòu)的變化,ELISA試劑盒分析仔鼠血清睪酮(T)、促卵泡素(FSH)及雌二醇(E2)水平,免疫組織化學方法檢測仔鼠睪丸或卵巢Bax、Bcl-2蛋白的表達,實時熒光定量PCR檢測仔鼠睪丸StAR、CYP11a或卵巢AMH、Kitlg mRNA表達。結(jié)果顯示,孕鼠暴露BPA極顯著增加了仔鼠死亡率(P0.01),顯著降低了仔鼠睪丸重(P0.05)。ELISA檢測結(jié)果表明,孕鼠暴露BPA極顯著降低了仔鼠T(♂)及FSH(♀)含量(P0.01),極顯著升高了仔鼠(♀)E2水平(P0.01)。HE染色結(jié)果顯示,隨BPA劑量增加,仔鼠睪丸組織損傷嚴重,間質(zhì)細胞減少;卵巢組織結(jié)構(gòu)隨BPA劑量增大,空泡逐漸增多,黃體顆粒數(shù)量逐漸減少。免疫組化結(jié)果顯示,孕鼠暴露BPA增加了仔鼠睪丸或卵巢組織中Bax陽性蛋白表達,減少了Bcl-2陽性蛋白表達,顯著降低了雄性仔鼠StAR mRNA表達量(P0.05);B、D組雄性仔鼠CYP11amRNA表達量極顯著降低(P0.01),而C組CYP11amRNA表達量極顯著升高(P0.01);C、D組雌鼠Kitlg mRNA表達極顯著降低(P0.01),AMH mRNA表達量顯著升高(P0.05)。本試驗結(jié)果表明,孕鼠妊娠期暴露不同劑量BPA增加了仔鼠死亡率,擾亂了生殖激素平衡和睪丸/卵巢中相關(guān)凋亡蛋白及生殖基因表達。
[Abstract]:In order to study the effect of bisphenol A on reproductive hormones and related genes in pregnant mice, 40 Kunming pregnant rats were randomly divided into 4 groups, 10 in each group. Group A was used as control group. Rats in group D were given BPA-50 ~ 500g / day for the whole gestational period (1 day of gestation to delivery), then the pregnant rats were given natural delivery, and the death of their offspring was observed and recorded. After sex maturation (56 days), the testis or ovaries were removed and weighed, and the changes of tissue structure of ovary or testis were observed by HE staining. The serum levels of testosterone (T), follicle stimulating hormone (FSH) and estradiol (E 2) were analyzed by Elisa kit. The expression of Bcl-2 protein in testis or ovary of newborn rats was detected by immunohistochemical method, and the expression of CYP11a in testis or PCR mRNA in ovary was detected by real-time fluorescence quantitative PCR. The results showed that BPA exposure significantly increased the infant mortality (P0.01) and significantly decreased the testis weight (P0.05). The results of Elisa showed that, BPA exposure significantly decreased the contents of T (P 0.01) and FSH (P 0.01), and significantly increased the level of E 2 (P 0.01). The results of HE staining showed that with the increase of BPA dose, the testicular tissue damage was serious and the interstitial cells decreased. With the increase of BPA dose, the vacuoles gradually increased and the number of luteal granules decreased. The results of immunohistochemistry showed that BPA increased the expression of Bax positive protein in testis or ovary, and decreased the expression of Bcl-2 positive protein in testis or ovary of pregnant rats. The expression of StAR mRNA in male rats was significantly decreased (P0.05), while the expression of CYP11amRNA in group C was significantly increased (P0.01). The expression of Kitlg mRNA in female rats was significantly decreased (P0.01) (P0.05). The results showed that exposure to different doses of BPA during pregnancy increased mortality, disturbed the balance of reproductive hormones and related apoptotic proteins and reproductive gene expression in testis / ovaries.
