維生素E拮抗雙酚A對卵巢顆粒細胞性激素分泌的抑制作用

【摘要】:目的·研究維生素E能否對雙酚A(BPA)的雌性性激素干擾效應起拮抗作用,探索拮抗作用的適宜濃度。方法·提取大鼠卵巢原代顆粒細胞。不同濃度BPA(0、0.01、0.1、1、10、100μmol/L)孵育48 h后,收集并用ELISA法測定細胞培養(yǎng)基中的雌二醇(E_2)及孕酮(P_4)濃度。采用健康女性卵泡液中的平均濃度(簡稱"人體平均濃度",5μmol/L)或高濃度(25μmol/L)維生素E(α-生育酚)單獨孵育或與BPA(10、100μmol/L)共同孵育顆粒細胞48 h后,測定培養(yǎng)液中性激素濃度。結果·10μmol/L及100μmol/L BPA可使大鼠原代卵巢顆粒細胞分泌性激素水平呈濃度依賴性降低(P0.05)。25μmol/L維生素E單獨孵育時可分別使E_2和P_4濃度顯著升高(44.89±15.18)%和(43.33±8.82)%(P0.05);維生素E(5μmol/L或25μmol/L)和BPA共同孵育時,性激素濃度恢復正常,與陰性對照組相比,差異無統(tǒng)計學意義(P0.05)。結論·人體平均濃度及高濃度維生素E均可拮抗BPA導致的性激素降低效應,而高濃度維生素E的改善效果相對顯著。
[Abstract]:Objective to study whether vitamin E can antagonize the female sex hormone interference effect of bisphenol A (BPA) and explore the appropriate concentration of the antagonism. Methods the primary granulosa cells of rat ovary were extracted. The concentrations of estradiol (ESP) and progesterone (PST4) in cell culture medium were determined by Elisa after incubated with different concentrations of BPA (0. 01 ~ 0. 1 ~ 0. 1 ~ 1 ~ (-1) 10 100 渭 mol 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) for 48 h. The average concentration of vitamin E (偽 -tocopherol) in healthy female follicular fluid (5 渭 mol / L for short) or high concentration (25 渭 mol / L) of vitamin E (偽 -tocopherol) was incubated alone or co-incubated with 10100 渭 mol / L of BPA for 48 h. The concentration of sex hormone in the medium was measured. Results 10 渭 mol / L and 100 渭 mol / L BPA decreased the level of sex hormones secreted by primary ovarian granulosa cells in a concentration-dependent manner (P0.05) .25 渭 mol / L vitamin E increased significantly (44.89 鹵15.18)% and (43.33 鹵8.82)% (P0.05), respectively, and vitamin E (5 渭 mol / L or 25 渭 mol / L) and BPA were incubated with BPA, respectively. Sex hormone concentration returned to normal, compared with the negative control group, the difference was not statistically significant (P0.05). Conclusion the average concentration of vitamin E and high concentration of vitamin E can antagonize the decrease of sex hormone induced by BPA, but the improvement of high concentration of vitamin E is relatively significant.
