【摘要】:目的: 鄰苯二甲酸二辛酯(DOP)作為增塑劑和添加劑等被廣泛應(yīng)用于塑料、農(nóng)藥、橡膠和化妝品等生產(chǎn)領(lǐng)域,由于其極易進(jìn)入到環(huán)境當(dāng)中且具有環(huán)境雌激素樣作用,可對(duì)環(huán)境和人體產(chǎn)生影響。氯仿作為氯化消毒的重要副產(chǎn)物,對(duì)人體同樣有害,但目前對(duì)于二者的聯(lián)合毒作用的報(bào)道較少。本實(shí)驗(yàn)用DOP和氯仿同時(shí)對(duì)小鼠進(jìn)行染毒,觀察其體重的變化,腦臟器系數(shù),腦的脂質(zhì)過氧化作用,腦組織酶活性以及對(duì)腦細(xì)胞的鈣離子濃度、線粒體膜電位的影響,來研究二者同時(shí)作用時(shí)對(duì)腦組織的損傷作用以及聯(lián)合毒性。 方法: 本實(shí)驗(yàn)采用聯(lián)合作用的分析方法—2×2析因分析進(jìn)行實(shí)驗(yàn)設(shè)計(jì)分組。實(shí)驗(yàn)設(shè)計(jì)包括DOP和氯仿兩個(gè)影響因素,均有暴露和未暴露兩個(gè)水平,分為不染毒的對(duì)照組、只暴露DOP染毒組、只暴露氯仿染毒組和同時(shí)暴露DOP和氯仿染毒組四種組合方式。 各組小鼠采用灌胃法染毒,隨機(jī)分為對(duì)照組(玉米油)、DOP單獨(dú)染毒組(1/10LD_(50),540mg/kg)、CF單獨(dú)染毒組(1/10LD_(50),112mg/kg)和DOP(1/10LD_(50),540mg/kg)+CF(1/10LD_(50),112mg/kg)聯(lián)合染毒組,雌雄各半,每組10只,各組均以玉米油作為溶劑。每天一次,連續(xù)染毒15天后處死小鼠,測定腦組織臟器系數(shù)、腦中神經(jīng)遞質(zhì)、腦的脂質(zhì)過氧化指標(biāo)、腦組織酶、腦中鈣離子濃度、線粒體膜電位。 結(jié)果: 1、DOP和氯仿聯(lián)合染毒對(duì)小鼠體重的影響 灌胃染毒15天后,雌性小鼠DOP染毒組體重高于對(duì)照組(P0.05),聯(lián)合組存在交互作用(P0.05);雄性小鼠氯仿組的體重低于對(duì)照組(P0.05)。 2、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦脂質(zhì)過氧化作用的影響 雌性小鼠DOP組腦組織中CAT的活力高于對(duì)照組(P0.05),聯(lián)合組存在交互作用(P0.05);雄性小鼠聯(lián)合組腦組織中CAT的活力存在交互作用(P0.05),氯仿組MDA含量高于對(duì)照組(P0.05),且聯(lián)合組存在交互作用(P0.05)。 3、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦神經(jīng)遞質(zhì)的影響 雌性小鼠腦中DOP組和氯仿組的5-HT、NE含量均低于對(duì)照組(P0.05),聯(lián)合染毒組均存在交互作用(P0.05);氯仿組DA含量低于對(duì)照組(P0.05),聯(lián)合染毒組存在交互作用(P0.05)。雄性小鼠DOP組的5-HT含量高于對(duì)照組(P0.05),氯仿組低于對(duì)照組(P0.05);聯(lián)合染毒組NE含量存在交互作用(P0.05);DOP組與氯仿組DA含量均高于對(duì)照組(P0.05),聯(lián)合染毒組存在交互作用(P0.05)。DOP和氯仿聯(lián)合染毒對(duì)小鼠腦神經(jīng)遞質(zhì)的影響除5-HT對(duì)照組外均存在性別差異(P0.05)。 4、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦Na~+-K~+-ATP酶與Ca~(2+)-Mg~(2+)-ATP酶的影響 雌性小鼠DOP組和氯仿組兩種酶的活力均高于對(duì)照組(P0.05),聯(lián)合組的Ca~(2+)-Mg~(2+)-ATP酶存在交互作用(P0.05);雄性小鼠氯仿組兩種酶的活力均高于對(duì)照組(P0.05),并且聯(lián)合組都存在交互作用(P0.05)。 5、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦LDH、、SDH、NOS、NO的影響 雌性小鼠氯仿組的LDH活性和SDH活性均高于對(duì)照組(P0.05),且聯(lián)合暴露組均存在交互作用(P0.05);DOP組的NO值低于對(duì)照組(P0.05),,聯(lián)合染毒組存在交互作用(P0.05)。雄性小鼠DOP組的LDH活性和SDH活性均低于對(duì)照組(P0.05),DOP組和氯仿組都存在交互作用(P0.05);氯仿組NOS活性高于對(duì)照組(P0.05);DOP組的NO值低于對(duì)照組(P0.05)。 6、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦線粒體膜電位與鈣離子濃度的影響 雌性小鼠DOP組和氯仿組的Ca~(2+)濃度低于對(duì)照組(P0.05),線粒體膜電位高于對(duì)照組(P0.05),聯(lián)合染毒組的Ca~(2+)濃度存在交互作用(P0.05);雄性氯仿組的線粒體膜電位高于對(duì)照組(P0.05),聯(lián)合暴露組存在交互作用(P0.05)。 結(jié)論: 1、同時(shí)接觸DOP和氯仿可以引起雌性小鼠CAT活性升高,雄性小鼠CAT活性降低,MDA含量升高。 2、雄性小鼠腦內(nèi)5-HT、NE、DA的含量升高,雌性降低,對(duì)神經(jīng)遞質(zhì)的作用在雌雄之間存在一定的性別差異。 3、雌性小鼠腦中LDH、SDH、NOS、NO、Ca~(2+)-Mg~(2+)-ATP酶的活力或含量均升高,雄性小鼠腦中LDH、SDH活性降低,Na~+-K~+-ATP酶與Ca~(2+)-Mg~(2+)-ATP酶活力升高。 4、雌性小鼠鈣離子濃度降低,雄雄小鼠線粒體膜電位升高。 5、DOP和氯仿聯(lián)合染毒對(duì)小鼠腦組織可產(chǎn)生交互作用。
[Abstract]:Purpose :

Dioctyl phthalate ( DOP ) is widely used as plasticizer and additive in the fields of plastics , pesticides , rubber and cosmetics . Because of its easy entry into the environment and has environmental estrogen - like effect , it can influence the environment and human body . Chloroform can be used as an important by - product for chlorination and disinfection , and it is harmful to human body . However , the effect of its body weight , brain organ coefficient , lipid peroxidation of brain , brain tissue enzyme activity and the effect of calcium ion concentration and mitochondrial membrane potential on brain cells are studied .

Method :

The experimental design consisted of two influencing factors : DOP and chloroform , both of which were exposed to and unexposed to two levels , divided into non - toxic control group , exposure to DOP staining group , exposure to chloroform staining group and exposure to four combinations of DOP and chloroform .

The rats were randomly divided into control group ( corn oil ) , DOP alone ( 1 / 10LD _ ( 50 ) , 540mg / kg ) , CF alone ( 1 / 10LD _ ( 50 ) , 112mg / kg ) and DOP ( 1 / 10LD _ ( 50 ) , 540mg / kg ) + CF ( 1 / 10LD _ ( 50 ) , 112mg / kg ) .

Results :

Effect of Combination of 1 , DOP and Chloroform on Weight of Mice

The weight of DOP in female mice was higher than that of control group ( P0.05 ) .
The weight of chloroform in male mice was lower than that of control group ( P0.05 ) .

Effects of combination of 2 , DOP and chloroform on lipid peroxidation in brain of mice

The activity of CAT in the brain tissue of female mice was higher than that in the control group ( P0.05 ) .
The activity of CAT in the brain tissue of male mice was higher than that in the control group ( P0.05 ) , and the content of MDA in chloroform group was higher than that in the control group ( P0.05 ) .
