青春期雙酚A暴露對小鼠認知功能和行為發(fā)育的影響

【摘要】:目的探討青春期(即出生后28-56天,PND28-56)雙酚A(Bispheno1A,BPA)暴露對ICR小鼠認知功能和行為的損害效應(yīng)及其是否有持續(xù)影響。 方法選取4周齡ICR小鼠,隨機均分為對照組、BPA(1、100mg/L)處理組。經(jīng)玻璃瓶飲水染毒4周后,于成年早期采用Morris水迷宮試驗來檢測空間學習和記憶能力,采用曠場試驗、黑白巷試驗、高架十字迷宮試驗來檢測探索和焦慮相關(guān)行為,采用攻擊行為實驗來檢測攻擊行為。停止染毒,飼養(yǎng)4周后,于成年期重復(fù)以上行為試驗,來觀察BPA暴露對小鼠認知功能、焦慮相關(guān)行為和攻擊行為的損害效應(yīng)是否具有持續(xù)影響。 結(jié)果 1.青春期和成年期階段均無疾病、感染和死亡現(xiàn)象,,體重的差異無統(tǒng)計學意義。 2.在Morris水迷宮實驗中,BPA(100mg/L)處理組成年早期雌性小鼠的潛伏期和游泳距離顯著延長(P 0.05),但雄鼠尚未觀測到有差異;在成年期,各指標沒有統(tǒng)計學差異。 3.在曠場實驗中,與對照組相比較,BPA(1mg/L)處理組成年早期雌鼠的潛伏期有統(tǒng)計學差異(P 0.05);BPA(1mg/L)處理組成年早期雌鼠和成年期的雄鼠站立次數(shù)較對照組顯著減少(P 0.05)。 4.在黑白巷實驗中,各觀測指標在成年早期差異沒有統(tǒng)計學差異;BPA(100mg/L)處理組成年期雌鼠的黑巷時間與對照組相比差異有統(tǒng)計學意義(P 0.05)。 5.在攻擊實驗,BPA(100mg/L)處理組成年早期雄鼠的攻擊潛伏期與對照組相比有差異(P 0.05);BPA(100mg/L)處理組成年早期和成年期雄鼠的接觸時間與對照組相比均有差異(P 0.05)。 結(jié)論青春期BPA暴露對小鼠空間學習和記憶功能的損害呈現(xiàn)顯著性別差異,對雌鼠損害效應(yīng)更為顯著;青春期BPA暴露對小鼠探索、焦慮和攻擊行為帶來持續(xù)性影響。
[Abstract]:Objective to investigate the effects of bisphenol A (BPA) exposure on cognitive function and behavior in ICR mice during puberty (28-56 days after birth). Methods four week old ICR mice were randomly divided into control group treated with BPA (1100mg / L). Four weeks after exposure to drinking water in glass bottles, Morris water maze test was used in early adulthood to test spatial learning and memory ability, open field test, black-and-white lane test, elevated cross maze test were used to detect behaviors related to exploration and anxiety. The attack behavior experiment was used to detect the attack behavior. After 4 weeks of feeding, the above behavioral tests were repeated in adult to observe whether BPA exposure had a lasting effect on cognitive function, anxiety related behavior and aggressive behavior in mice. Result 1. There was no disease, infection and death, and there was no significant difference in body weight between puberty and adulthood. In Morris water maze test, BPA (100 mg / L) significantly prolonged the latency and swimming distance of female mice in the early years of composition (P 0.05), but no significant difference was found in male mice, but there was no statistical difference among the indexes in adulthood. In the open field experiment, compared with the control group, there was a significant difference (P 0.05) in the incubation period of the female rats treated with BPA (1 mg / L) in the early stage of the constituent year (P 0.05), and the number of standing times of the male rats in the early stage of the composition year and the adult period was significantly decreased compared with the control group (P 0.05) (P 0.05). In the black-and-white lane experiment, there was no statistical difference between the observed indexes in the early adulthood. The black lane time of the female rats made up of BPA (100 mg / L) was significantly different from that of the control group (P 0.05). There was a significant difference in the incubation period between the male and the control group after BPA (100 mg / L) treatment (P 0.05). The exposure time of BPA (100 mg / L) was significantly different from that of the control group (P 0.05), and that of BPA (100 mg / L) was higher than that of the control group (P < 0.05). Conclusion the impairment of spatial learning and memory in mice exposed to pubertal BPA is significantly different from that of female mice, and the effects of puberty BPA exposure on exploration, anxiety and aggressive behavior in mice are persistent.
