【摘要】：目的探討亞硫酸鈉對(duì)人正常二倍體肝細(xì)胞(HL-7702)的損傷作用及程序性壞死相關(guān)蛋白受體相互作用蛋白1(RIP1)表達(dá)的影響。方法將處于對(duì)數(shù)生長(zhǎng)期的HL-7702細(xì)胞分別暴露于含終濃度為0(對(duì)照)、0.01、0.1、1、10 mmol/L亞硫酸鈉的培養(yǎng)基中培養(yǎng)2、4、12、24、48 h。采用CCK-8法檢測(cè)細(xì)胞活性,采用全自動(dòng)生化分析儀檢測(cè)細(xì)胞上清液中丙氨酸氨基轉(zhuǎn)移酶(ALT)、天冬氨酸氨基轉(zhuǎn)移酶(AST)以及乳酸脫氫酶(LDH)的活力,采用Western Blot法檢測(cè)細(xì)胞RIP1蛋白的表達(dá)水平。結(jié)果與對(duì)照組相比,1、10 mmol/L亞硫酸鈉染毒不同時(shí)間的HL-7702細(xì)胞存活率降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05);且隨著亞硫酸鈉染毒濃度的升高,HL-7702細(xì)胞的存活率呈下降趨勢(shì)。與對(duì)照組相比,僅10 mmol/L亞硫酸鈉染毒24、48 h HL-7702細(xì)胞上清液中的ALT活力較高;1 mmol/L亞硫酸鈉染毒4、24 h及10 mmol/L亞硫酸鈉染毒2、4 h HL-7702細(xì)胞上清液中的AST活力較低,而10 mmol/L亞硫酸鈉染毒24、48 h HL-7702細(xì)胞上清液中的AST活力較高;10 mmol/L亞硫酸鈉染毒2、4 h HL-7702細(xì)胞上清液中的LDH活力較低,而染毒24、48 h HL-7702細(xì)胞上清液中的LDH活力較高,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。與對(duì)照組相比,各濃度亞硫酸鈉染毒組HL-7702細(xì)胞中RIP1蛋白的表達(dá)水平均較高,差異多有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論程序性壞死可能參與了亞硫酸鈉引起的肝細(xì)胞死亡,但其可能機(jī)制還有待于進(jìn)一步研究。
[Abstract]:Objective to investigate the effects of sodium sulfite on the damage of human diploid hepatocytes HL-7702 and the expression of programmed necrosis associated protein receptor interaction protein 1 (RIP1). Methods HL-7702 cells at logarithmic growth stage were exposed to a culture medium containing 0 (0. 01) (control 0. 01) and 10 mmol 路L ~ (-1) sodium sulfite, respectively. The activity of cells was detected by CCK-8 method, and the activities of alanine aminotransferase (alt), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) in supernatant of cells were detected by automatic biochemical analyzer. The expression of RIP1 protein was detected by Western blot. Results compared with the control group, the survival rate of HL-7702 cells exposed to 10 mmol / L sodium sulfite decreased significantly (P 0.05), and the survival rate of HL-7702 cells decreased with the increase of sodium sulfite concentration. Compared with the control group, the alt activity in the supernatant of HL-7702 cells exposed to 10 mmol / L sodium sulfite for 24 h or 48 h was higher than that in the supernatant of HL-7702 cells exposed to 10 mmol / L sodium sulfite for 24 h and 10 mmol / L sodium sulfite for 24 h and 2 h, respectively, and the AST activity in the supernatant of HL-7702 cells was lower than that in the supernatant of HL-7702 cells. The activity of AST in the supernatant of HL-7702 cells was higher than that in the supernatant of HL-7702 cells exposed to 10 mmol / L sodium sulfite for 24 h for 48 h. The activity of LDH in the supernatant of HL-7702 cells was lower than that in the supernatant of HL-7702 cells treated with 10 mmol / L sodium sulfite for 2 h or 4 h, but the activity of LDH in the supernatant of HL-7702 cells was higher than that in the supernatant of HL-7702 cells at 24 h after exposure to 24 mmol / L sodium sulfite. The difference was significant (P 0.05). Compared with the control group, the expression of RIP1 protein in HL-7702 cells exposed to sodium sulfite at different concentrations was higher than that in the control group, and the difference was statistically significant (P 0.05). Conclusion programmed necrosis may be involved in the hepatic cell death induced by sodium sulfite, but the possible mechanism remains to be further studied.
