【摘要】：目的探討1.0μmol/L亞砷酸鈉慢性染毒對(duì)人支氣管上皮細(xì)胞(HBE)和人角質(zhì)形成細(xì)胞(Ha Ca T)體外增殖的改變及其調(diào)控機(jī)制。方法以體外培養(yǎng)的HBE和HaCaT細(xì)胞惡性轉(zhuǎn)化模型為研究對(duì)象,應(yīng)用MTT法檢測(cè)細(xì)胞增殖情況,采用流式細(xì)胞儀檢測(cè)細(xì)胞周期改變,利用Western blot檢測(cè)細(xì)胞周期相關(guān)蛋白cyclin E、cyclin D1、cylin A蛋白的表達(dá)。結(jié)果染砷組HBE細(xì)胞36代和43代、HaCaT細(xì)胞28代和35代相對(duì)增殖率均高于傳代對(duì)照組(P0.01和P0.05);細(xì)胞周期G1期細(xì)胞比例下降(P0.05),S期細(xì)胞比例上升(P0.05);此外,細(xì)胞周期相關(guān)蛋白cyclin E表達(dá)升高(P0.05),并在晚期持續(xù)性高表達(dá)。結(jié)論低劑量亞砷酸鈉可通過上調(diào)HBE和Ha Ca T細(xì)胞中cyclin E的表達(dá)從而使細(xì)胞從G1期逃逸至S期,加速細(xì)胞增殖,最終導(dǎo)致細(xì)胞發(fā)生惡性轉(zhuǎn)化。
[Abstract]:Objective to investigate the effects of 1.0 渭 mol / L sodium arsenite on the proliferation of human bronchial epithelial cells (HBE) and human keratinocytes (HA-Ca T) in vitro. Methods the malignant transformation model of HBE and HaCaT cells in vitro was used to detect the proliferation of HBE and HaCaT cells, and the cell cycle was detected by flow cytometry. Western blot was used to detect the expression of cyclin D 1 cylin A protein in cell cycle associated protein cyclin. Results the relative proliferation rate of HaCaT cells at passage 28 and 35 in arsenic exposed group was higher than that in control group (P0.01 and P0.05), while the percentage of cells in G1 phase of cell cycle decreased in P0.05 / S phase. The expression of cell cycle associated protein (cyclin E) was increased in P0. 05 and continued to be high in late stage. Conclusion low dose sodium arsenite can up-regulate the expression of cyclin E in HBE and Ha Ca T cells, so that the cells escape from G1 phase to S phase, accelerate cell proliferation, and eventually lead to malignant transformation.
