【摘要】：研究卷煙煙氣暴露對大鼠學(xué)習(xí)記憶功能的影響,并探討腦源性神經(jīng)營養(yǎng)因子(BDNF)的作用。將雄性SD大鼠隨機(jī)分為對照組、低劑量組(染毒10min)、中劑量組(染毒20min)及高劑量組(染毒40min),置于氣體染毒箱內(nèi)行被動吸煙,分別于煙氣暴露后的30d、90d、180d進(jìn)行Morris水迷官實驗,檢測大鼠學(xué)習(xí)記憶功能,觀察指標(biāo)包括：動物體重,外周血血細(xì)胞參數(shù),血清中同型半胱氨酸(Hcy)含量,HE染色觀察海馬區(qū)病理,免疫組化方法觀察海馬BDNF表達(dá)變化。 大鼠卷煙煙氣暴露后,所檢測各個指標(biāo)與正常對照組相比較,結(jié)果如下： (1)大鼠體重增長明顯下降； (2)白細(xì)胞數(shù)先增加后減少,紅細(xì)胞數(shù)則呈先減少后增加的趨勢； (3)染毒30d,煙氣暴露組潛伏期均顯著低于對照組(P0.05)；染毒90d,中劑量煙氣暴露組潛伏期顯著低于對照組(P0.05)；染毒180d,煙氣暴露組潛伏期顯著高于對照組(P0.05)。同組間隨時間的增加,對照組表現(xiàn)為學(xué)習(xí)記憶功能逐漸增強(qiáng),低劑量組為先降低后增強(qiáng),中劑量組為增強(qiáng),高劑量組無明顯變化。 (4)HE染色結(jié)果表明卷煙煙氣暴露30d、90d、180d大鼠大腦海馬區(qū)無明顯變化； (5)染毒30d至90d,低劑量組血清中Hcy含量增加,中劑量組和高劑量組含量下降；染毒180d,染毒組血清中Hcy含量均明顯高于對照組。 (6)；染毒30d,煙氣暴露組潛伏期BDNF表達(dá)顯著高于對照組(P0.05)；染毒90d,中劑量煙氣暴露組BDNF表達(dá)顯著高于對照組(P0.05)；染毒180d,煙氣暴露組BDNF表達(dá)顯著高于對照組(P0.05)。 短期煙氣暴露可提高大鼠學(xué)習(xí)記憶功能,隨染毒時間增長,學(xué)習(xí)記憶功能下降。該作用與海馬BDNF的表達(dá)密切相關(guān)。
[Abstract]:To study the effects of cigarette smoke exposure on learning and memory function in rats and to explore the role of brain-derived neurotrophic factor (BDNF). Male Sprague-Dawley rats were randomly divided into control group, low dose group (10 min exposure), middle dose group (20 min exposure) and high dose group (40 min exposure). The learning and memory function of rats was measured. The indexes were as follows: body weight, peripheral blood cell parameters, serum homocysteine (Hcyl) content and HE staining to observe the pathological changes of hippocampus and the expression of BDNF in hippocampus by immunohistochemical method. Compared with the normal control group, the results were as follows: (1) the weight gain of rats decreased significantly, and the white blood cell count increased first and then decreased. The number of red blood cells decreased first and then increased, the incubation period of the smoke exposure group was significantly lower than that of the control group for 30 days, and the incubation period of the middle dose smoke exposure group was significantly lower than that of the control group for 90 days. The incubation period of the exposed group was significantly higher than that of the control group for 180 days. With the increase of time, the learning and memory function of the control group was gradually enhanced, that of the low dose group was decreased first and then increased, and that of the middle dose group was enhanced. The results of HE staining showed that there was no significant change in hippocampal area of rats exposed to cigarette smoke for 30 days or 90 days after exposure to cigarette smoke, while the content of Hcy in serum of low dose group increased from 30 days to 90 days after exposure to cigarette smoke. The contents of serum Hcy in the medium dose group and high dose group were significantly higher than those in the control group at 180 days after exposure, and the expression of BDNF in the incubation period of the smoke exposure group was significantly higher than that in the control group at 30 days after exposure. The expression of BDNF in the exposure group was significantly higher than that in the control group for 90 days, and the expression of BDNF in the smoke exposure group was significantly higher than that in the control group at 180 days. Short-term smoke exposure could improve the learning and memory function of rats, and the learning and memory function decreased with the increase of exposure time. This effect is closely related to the expression of BDNF in hippocampus.
