普洱茶茶色素對(duì)HepG2細(xì)胞脂質(zhì)代謝的影響及作用機(jī)理

【摘要】:以正常培養(yǎng)和油酸誘導(dǎo)培養(yǎng)的人肝癌細(xì)胞系(human hepatocellular liver carcinoma cell line,Hep G2)細(xì)胞為模型,通過(guò)測(cè)定普洱茶茶色素對(duì)Hep G2細(xì)胞內(nèi)甘油三酯(triglyceride,TG)和總膽固醇(total cholesterol,TC)含量,細(xì)胞中脂肪酸合成酶(fatty acid synthase,FAS)、固醇調(diào)節(jié)元件結(jié)合蛋白1c(sterol regulatory element binding protein 1c,SREBP-1c)、三磷酸腺苷結(jié)合轉(zhuǎn)運(yùn)子A1(ATP binding cassette transporter A1,ABCA1)、膽固醇7α-羥化酶(cholesterol 7α-hydroxylase,CYP7A1)的轉(zhuǎn)錄水平,磷酸化腺苷酸活化蛋白激酶(phospho-AMPactivated protein kinase,p-AMPK)的蛋白表達(dá)水平研究普洱茶茶色素的減肥降脂作用機(jī)制。結(jié)果顯示,普洱茶茶色素能明顯降低油酸誘導(dǎo)的Hep G2細(xì)胞模型中TG和TC含量,作用程度依賴(lài)于普洱茶的作用質(zhì)量濃度。但對(duì)正常培養(yǎng)的Hep G2的TG、TC作用影響不顯著。經(jīng)過(guò)普洱茶茶色素作用油酸誘導(dǎo)Hep G2細(xì)胞24 h后,能顯著下調(diào)細(xì)胞的FAS和SREBP-1c的m RNA表達(dá)水平(P0.05),顯著上調(diào)ABCA1的轉(zhuǎn)錄水平(P0.05),且使CYP7A1的轉(zhuǎn)錄水平呈上升趨勢(shì),并顯著上調(diào)p-AMPK蛋白的表達(dá)量(P0.05)。因此,普洱茶茶色素可通過(guò)調(diào)控上述調(diào)控因子和酶的表達(dá)而改善油酸誘導(dǎo)下Hep G2細(xì)胞的脂質(zhì)代謝水平。
[Abstract]:The normal and oleic acid-induced human hepatocellular liver carcinoma cell lineage Hep G2 (Hep G2) cells were used as models. The contents of triglyceride (TG) and total cholesterol (TC) in Hep G2 cells treated with tea pigment from Pu'er tea were measured. The transcriptional levels of fatty acid synthase (acid synthase), steroid regulatory element binding protein (1c(sterol regulatory element binding protein 1), adenosine triphosphate binding transporter (A1(ATP binding cassette transporter A1) ABCA1, cholesterol 7 偽 -hydroxylase1 (CYP7A1) and fatty acid synthase (acid synthase) were detected. Protein expression of phosphorylated adenylate activated protein kinase phospho-AMPactivated protein kinase- p-AMPK study on the mechanism of reducing fat and reducing fat of tea pigment in Pu'er tea. The results showed that the content of TG and TC in Hep G2 cells induced by oleic acid was significantly decreased by tea pigment, which depended on the action concentration of Pu'er tea. However, the effect of Hep G2 on TC was not significant. After induced by oleic acid for 24 h, the expression level of m RNA of FAS and SREBP-1c in Hep G2 cells was significantly down-regulated, and the transcription level of ABCA1 was upregulated, and the transcription level of CYP7A1 was on the rise. The expression of p-AMPK protein was significantly up-regulated (P 0.05). Therefore, tea pigment can improve the lipid metabolism of Hep G2 cells induced by oleic acid by regulating the expression of these regulatory factors and enzymes.
