氟染毒對大鼠皮膚成纖維細胞中SOST及DKK1蛋白表達的影響

【摘要】:[目的]觀察氟染毒對體外培養(yǎng)新生大鼠皮膚成纖維細胞硬骨素(sclerostin,SOST)和Dickkopf-1(DKK1)蛋白表達的影響。[方法]采用原代培養(yǎng)方法,將大鼠皮膚成纖維細胞分為對照組(0 mg/L)和7個染氟劑量組(0.000 1、0.001、0.01、0.1、1、10、20 mg/L),分別在4個染氟時間段(24、48、72、96 h)收集細胞培養(yǎng)上清液,應用ELISA法測定SOST和DKK1蛋白表達水平。[結果]與染氟0 mg/L組相比,SOST表達量在染氟48 h的0.000 1、0.001、0.01 mg/L組,染氟72 h的1、10 mg/L組及染氟96 h的0.001、0.01、0.1、1 mg/L組均明顯下降(P0.05)。與染氟24 h組相比,SOST表達量在染氟48 h的0.01 mg/L組,染氟96 h的0.001、0.01、0.1、1、10 mg/L組均明顯下降(P0.05)。與染氟0 mg/L組相比,DKK1表達量在染氟48 h的0.001 mg/L組,染氟72 h的0.01、0.1、1、10、20 mg/L組及染氟96 h的0.01、0.1、1、10、20 mg/L組均明顯下降(P0.05)。與染氟24 h組相比,DKK1表達量在染氟48 h的0.000 1、0.001、0.01 mg/L組,染氟72 h的0.001、0.01、1、10、20 mg/L及染氟96 h的0.01、0.1、1、20 mg/L組均明顯下降(P0.05)。[結論]氟可導致大鼠皮膚成纖維細胞SOST和DKK1蛋白表達下調。
[Abstract]:[objective] to observe the effect of fluoride exposure on the expression of osteostatin (sclerostinine) and Dickkopf-1 (DKK1) protein in cultured skin fibroblasts of neonatal rats in vitro. [methods] Rat skin fibroblasts were divided into two groups: control group (0 mg / L) and fluorine-exposed group (0.000 1g / L). The supernatants were collected from rat skin fibroblasts at 4 fluorosis time periods (24 ~ 448 ~ 7296 h). The expression of SOST and DKK1 protein was determined by ELISA assay. [results] compared with the fluorine 0 mg/L group, the expression of sst was significantly decreased in the 0.000 1U 0.001 mg/L, 72 h 10 mg/L and 96 h fluorinated 0. 01 mg/L groups, respectively (P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05). Compared with the 24 h fluoride exposure group, the expression of sst was significantly decreased in the 0. 01 mg/L group at 48 h, and the 0. 001 0. 01 + 0. 1 + 1 + 10 mg/L group at 96 h after fluoride exposure (P 0. 05, P 0. 05, P 0. 05, P 0. 05, P 0. 05). Compared with the 0 mg/L group, the expression of DKK1 in the 0.001 mg/L group at 48 h, the 0. 01 ~ 0. 1 + 10 ~ 10 ~ 20 mg/L group in 72 h fluoride exposure group and the 0. 01 0. 1 1 + 10 ~ 10 ~ 20 mg/L group in 96 h fluoride group were significantly decreased (P 0. 05). Compared with the 24 h group, the expression of DKK1 was significantly decreased in the 0.000 1U 0. 001 mg/L group at 48 h, the 0. 001 0. 01 0. 01 mg/L in 72 h fluoride exposure group, and the 0. 01 + 0. 1 0. 01 mg/L group in 96 h fluoride exposure group. The expression of DKK1 protein was significantly decreased in the 72 h fluorine exposure group (P 0. 05) compared with the 24 h fluorine exposure group (P 0. 05). [conclusion] fluoride can induce down-regulation of SOST and DKK1 protein expression in rat skin fibroblasts.
