鹽酸柯諾拉贊對新西蘭白兔的胚胎-胎仔發(fā)育毒性及其毒代研究(英文)

目的研究鹽酸柯諾拉贊(KFP-H008)對新西蘭白兔的胚胎-胎仔發(fā)育毒性。方法將鹽酸柯諾拉贊(KFP-H008)5,15和50 mg·kg-1于新西蘭白兔妊娠第6~18天(GD 6~18,致畸敏感期)ig給予孕兔,同時設(shè)立溶媒對照組和陽性對照組(環(huán)磷酰胺10 mg·kg-1,iv),觀察并記錄母體在孕期體質(zhì)量和耗食量。在GD29對孕兔實施安樂死,記錄妊娠子宮、胎盤總重、黃體數(shù)、著床數(shù)、活胎數(shù)、死胎和吸收胎數(shù)等。觀察并記錄活胎的外觀、身長、體質(zhì)量,以及其骨骼和內(nèi)臟的發(fā)育情況。同時檢測鹽酸柯諾拉贊在衛(wèi)星組孕兔血漿中原形和代謝產(chǎn)物的濃度及其在孕兔和胎兔體內(nèi)的組織分布。結(jié)果從GD 13開始,KFP-H008 15和50 mg·kg-1組孕兔體質(zhì)量和耗食量低于對照組(P<0.05)。同時上述各組胎兔的頂臀長和體質(zhì)量均明顯減少(P<0.05),且可見骨骼畸形或變異。KFP-H008單次給藥后被迅速代謝,在母體血漿中幾乎檢測不到。重復(fù)多次給藥后,KFP-H008 50 mg·kg-1組母體和胎仔組織中...
