一種萘啶衍生物3x通過非Caspase凋亡途徑誘導(dǎo)A375細(xì)胞死亡(英文)

萘啶化合物是一類具有多種生理學(xué)特性的藥物結(jié)構(gòu)單元,在醫(yī)學(xué)、藥學(xué)、生理學(xué)等諸多領(lǐng)域都有著廣泛的應(yīng)用.一種新的化合物(10-甲氧基-1,2,3,4-四氫苯并[g][1,3]二氮雜卓[1,2-a][1,8]萘并吡啶-6-基)(4-甲基環(huán)己-2,4-二烯-1-基)甲酮(命名為3x),經(jīng)噻唑藍(lán)四唑溴化銨(MTT)檢測,在人惡性黑色素瘤細(xì)胞株A375中顯示出良好的抗癌活性和選擇性.通過免疫印跡法的驗證,我們發(fā)現(xiàn)盡管3x可以明顯降低XIAP的表達(dá)量,但其并不通過激活Caspase-3,-8,-9等蛋白來誘導(dǎo)A375細(xì)胞死亡,而是極有可能通過引起p53的增加以及p53的磷酸化來致使Bax、PUMA、AIF的表達(dá)量增加,最終導(dǎo)致A375細(xì)胞死亡.
