化合物NS5806對(duì)Kv4通道的調(diào)節(jié)作用及其分子機(jī)制

Kv4通道是構(gòu)成快速激活和失活的瞬時(shí)外向電流的主要亞型,包括神經(jīng)元A型鉀電流(ISA)和心肌細(xì)胞瞬時(shí)外向鉀電流(Ito)。由于獨(dú)特的動(dòng)力學(xué)特征,Kv4通道在控制神經(jīng)元興奮和心肌動(dòng)作電位(AP)形態(tài)中發(fā)揮重要作用。在許多外周和中樞神經(jīng)元中發(fā)現(xiàn)的A型鉀電流(ISA),在決定動(dòng)作電位波形,重復(fù)放電特性并且在調(diào)節(jié)突觸傳遞和突觸可塑性方面起決定因素。另一方面,瞬時(shí)外向鉀電流(Ito)是大型哺乳動(dòng)物心臟動(dòng)作電位的1期復(fù)極化電流,決定動(dòng)作電位平臺(tái)期的高度,而在成年嚙齒類(lèi)動(dòng)物的心臟中Ito是主要的復(fù)極化電流。在許多病理生理?xiàng)l件下如:心臟肥大和心力衰竭(HF)都發(fā)現(xiàn)Ito密度的降低并伴隨有APD延長(zhǎng),從而導(dǎo)致心律失常。越來(lái)越多的證據(jù)表明,心臟衰竭過(guò)程中Ito減少不僅僅是繼發(fā)的改變,它還可能是HF發(fā)展的中介或啟動(dòng)因素。因此,提高Ito密度有望成為一種治療心臟肥大和心力衰竭等心臟疾病的策略。人們普遍認(rèn)為,根據(jù)不同的復(fù)活動(dòng)力學(xué)特征將I<...
