黑根霉胞外多糖促進(jìn)U87MG細(xì)胞自噬及其分子機(jī)制研究

自噬是細(xì)胞質(zhì)組分利用溶酶體降解的主要途徑。自噬過(guò)程發(fā)生異常與多種人類疾病相關(guān),如癌癥、心肌病等。目前,通過(guò)藥物靶向調(diào)節(jié)細(xì)胞自噬活性以達(dá)到抗腫瘤目的,已經(jīng)成為癌癥治療的新策略。多糖調(diào)節(jié)自噬活性的研究逐漸引起人們的關(guān)注,并且已取得一定得進(jìn)展。黑根霉胞外多糖(exopolysaccharides from Rhizopus nigricans,EPS1-1)是從黑根霉菌的發(fā)酵液中分離得到的一種均一的雜多糖。前期研究發(fā)現(xiàn),EPS1-1具有免疫增強(qiáng)活性以及良好的抗腫瘤活性,能夠誘導(dǎo)腫瘤細(xì)胞凋亡,對(duì)S180和CT26荷瘤小鼠以及AOM/DSS誘導(dǎo)的結(jié)腸癌小鼠腫瘤的生長(zhǎng)也有明顯的抑制作用。但是,EPS1-1對(duì)腫瘤生長(zhǎng)的抑制作用是否與調(diào)控自噬活性有關(guān)目前是未知的。因此,本文研究了EPS1-1對(duì)U87MG細(xì)胞自噬的影響及其作用機(jī)制,并對(duì)EPS1-1影響自噬和其與凋亡間的關(guān)系進(jìn)行了初步探討。通過(guò)MTT法和細(xì)胞克隆形成實(shí)驗(yàn)研究發(fā)現(xiàn)EPS1-1可以有效抑制U87MG細(xì)胞生長(zhǎng)和增殖。電鏡觀察發(fā)現(xiàn)0.4mg/ml的EPS1-1處理48h后,U87MG細(xì)胞內(nèi)產(chǎn)生了自噬溶酶體。自噬小體膜結(jié)合形式的LC3(LC3-Ⅱ)...
