行為學(xué)訓(xùn)練對局灶性腦缺血大鼠內(nèi)源性神經(jīng)干細胞增殖及PS1蛋白表達的影響

【摘要】:目的檢測早老素(Presenilinl,PS1)在局灶性腦缺血大鼠海馬組織神經(jīng)干細胞中的表達及穿梭箱訓(xùn)練的干預(yù)作用,探討影響神經(jīng)干細胞增殖分化的分子調(diào)控機制。方法采用大腦中動脈栓塞(MCAO)制作局灶性腦缺血大鼠模型,用免疫組織化學(xué)方法檢測海馬神經(jīng)干細胞Brd U、PS1的表達及穿梭箱訓(xùn)練的干預(yù)作用。結(jié)果隨著腦缺血再灌注3 d缺血海馬神經(jīng)元Brd U陽性細胞明顯增多,7 d達高峰。PS1反應(yīng)產(chǎn)物腦缺血再灌注7 d較正常組增多,隨缺血再灌注時間的進一步延長逐漸減少,14 d后持續(xù)低水平表達。穿梭箱訓(xùn)練后Brd U、PS1蛋白的表達明顯增加。結(jié)論穿梭箱訓(xùn)練促進神經(jīng)干細胞的增殖及缺血腦組織PS1的表達,提示穿梭箱訓(xùn)練促進神經(jīng)干細胞增殖作用可能由PS1信號介導(dǎo)。
[Abstract]:Objective to detect the expression of Presenilinl,PS1 in hippocampal neural stem cells of rats with focal cerebral ischemia and the intervention effect of shuttle box training, and to explore the molecular regulation mechanism of neural stem cells proliferation and differentiation. Methods the rat model of focal cerebral ischemia was established by middle cerebral artery embolism (MCAO). The expression of Brd U and PS1 in hippocampal neural stem cells and the intervention effect of shuttle box training were detected by immunohistochemistry. Results the number of Brd U positive cells in ischemic hippocampal neurons increased significantly with 3 days of cerebral ischemia and reperfusion, and reached the peak at 7 days. PS1 reaction products increased after 7 days of cerebral ischemia and reperfusion, and decreased gradually with the further prolongation of ischemia and reperfusion time, and continued to low expression after 14 days. The expression of PS1 protein increased significantly after shuttle box training. Conclusion shuttle box training promotes the proliferation of neural stem cells and the expression of PS1 in ischemic brain tissue, suggesting that the effect of shuttle box training on the proliferation of neural stem cells may be mediated by PS1 signal.
