HMGB1抗體對刀豆蛋白A引起小鼠肝損傷的保護作用

【摘要】:目的研究高遷移率族蛋白1(HMGB1)抗體對刀豆蛋白A(ConA)引起小鼠肝損傷的保護作用。方法將健康無污染雄性Balb/c小鼠分成空白對照組(注射生理鹽水)、模型組(注射ConA)和實驗組(注射ConA+HMGB1抗體),3組小鼠注射藥物6 h后摘眼球取血,所獲血液離心后取血清檢測谷丙轉(zhuǎn)氨酶(ALT)和HMGB1,留取肝臟組織進行HE染色、Tunel、免疫熒光等檢測。結(jié)果實驗組小鼠肝組織病理炎癥反應(yīng)輕于模型組。小鼠血清中ALT、HMGB1:空白對照組分別為(52.00±8.34)U/L、(7.54±0.53)ng/mL,模型組分別為(5 551.50±1 445.74)U/L、(18.06±1.65)ng/mL,實驗組分別為(1 977.40±654.89)U/L、(10.77±0.71)ng/mL;肝組織中HMGB1 mRNA、HMGB1表達量(相對值):空白對照組分別為1.886±0.253、0.086±0.028,模型組分別為4.718±0.341、0.268±0.043,實驗組分別為3.005±0.331、0.116±0.008;實驗組小鼠血清中ALT、HMGB1,以及肝組織中HMGB1 mRNA、HMGB1表達量均低于模型組(均P0.001)。肝組織細胞凋亡、肝細胞內(nèi)HMGB1遷移(標(biāo)準(zhǔn)化):空白對照組均為1±0,模型組分別為4.67±0.33、4.50±0.22,實驗組分別為2.67±0.21、2.33±0.21;實驗組小鼠肝組織細胞凋亡、肝細胞內(nèi)HMGB1遷移均低于模型組(均P0.001)。結(jié)論 HMGB1抗體能改善肝組織的病理損傷,可以保護ConA引起的小鼠肝損傷。

After centrifugation, serum samples were taken to detect glutamic-pyruvic transaminase (ALT) and HMGB1, for HE staining and Tunel, immunofluorescence.
