【摘要】：目的建立測定人血漿中左舒必利的高效液相色譜-質(zhì)譜聯(lián)用(HPLC-MS/MS)方法。方法以硫必利為內(nèi)標,血漿樣品經(jīng)甲醇沉淀蛋白提取分離,色譜柱為Xterra~汶RP18(4.6 mm×150 mm,5μm),流動相為乙腈-10 mmol·L~(-1)醋酸銨(含0.2%甲酸)水溶液(10:90),流速為1.0mL·min~(-1),柱溫為30℃,選用電噴霧離子源(ESI)在正離子電離模式下,采用多反應監(jiān)測(MRM)的質(zhì)譜掃描方式。結(jié)果左舒必利標準曲線方程y=1.75×10~(-2)x+2.93×10~(-3)(n=3,r=0.999 3),線性范圍1.00~600.00μg·L~(-1),定量下限為1.00μg·L~(-1),提取回收率在92.12%~98.53%,批內(nèi)、批間RSD均小于4.61%。結(jié)論本方法專屬性強,靈敏度高,操作簡便、快速、準確,適用于血漿中左舒必利濃度的測定。
[Abstract]:Objective to establish a high performance liquid chromatography-mass spectrometry (HPLC-MS/MS) method for the determination of levosulpiride in human plasma.
