【摘要】:目的:采用響應(yīng)面法優(yōu)選出制備穿膜肽(CPP)修飾紫杉醇和他莫昔芬脂質(zhì)體的最佳處方。方法:采用薄膜分散-硫酸銨梯度法制備CPP修飾紫杉醇和他莫昔芬脂質(zhì)體,以磷脂/膽固醇(摩爾比),磷脂/聚乙二醇-二硬脂酰磷脂酰乙醇胺(摩爾比),磷脂/紫杉醇(摩爾比)為自變量,以修飾紫杉醇和他莫昔芬包封率的綜合評(píng)分為因變量進(jìn)行二次多項(xiàng)擬合。結(jié)果:脂質(zhì)體的最優(yōu)處方為磷脂/膽固醇=50:1,磷脂/PEG 2000-DSPE=5.5:1,磷脂/紫杉醇=50:1,脂質(zhì)體中紫杉醇和他莫昔芬含量的綜合評(píng)分為95.48%。結(jié)論:星點(diǎn)設(shè)計(jì)-響應(yīng)面法優(yōu)選出了CPP修飾紫杉醇和他莫昔芬脂質(zhì)體的最佳處方。
[Abstract]:Aim: to optimize the preparation of paclitaxel and tamoxifen liposomes modified by transmembrane peptide (CPP) by response surface method. Methods: CPP modified paclitaxel and tamoxifen liposomes were prepared by thin film dispersion ammonium sulfate gradient method with phospholipid / cholesterol (molar ratio), phospholipid / polyethylene glycol distearyl phosphatidylethanolamine (molar ratio). Phospholipid / paclitaxel (molar ratio) was used as independent variable, and the comprehensive score of entrapment efficiency of modified paclitaxel and tamoxifen was used as dependent variable for quadratic multivariate fitting.
