固相萃取-HPLC法測定人血清中10-羥基卡馬西平的濃度

【摘要】:目的:建立測定人血清中10-羥基卡馬西平(MHD)濃度的方法,促進臨床個體化用藥和相關藥動學研究。方法:血清樣品采用固相萃取,用高效液相色譜法進樣測定。色譜柱為Venusil C18,流動相為甲醇-水(45∶55),流速為0.8 ml/min,紫外檢測波長為230 nm。結果:本試驗中內(nèi)源性物質(zhì)不干擾測定,MHD血藥濃度在2.49~39.82 mg/L范圍內(nèi)線性關系良好(r=0.999 8),檢測限為0.4 mg/L;高、中、低(2.49、19.91、39.82 mg/L)質(zhì)量濃度平均提取回收率為97.2%~101.2%,平均方法回收率為98.3%~100.8%,日內(nèi)和日間RSD均10%。結論:該方法快速、穩(wěn)定、準確,適用于MHD的血藥濃度測定和相關藥動學研究。
