【摘要】：為了評(píng)價(jià)仿制生物制品(follow-on biologics)的生物類(lèi)似性,美國(guó)食品藥品監(jiān)督管理局(FDA)在2012年2月頒布了3個(gè)指導(dǎo)原則草案。在有關(guān)"科學(xué)考量"(scientific consideration)的指導(dǎo)原則草案中,FDA要求產(chǎn)品的相似性必須在安全性、純度和有效性等各特征區(qū)中具有"證據(jù)鏈完備性"(totality-of-the-evidence)。然而,即使在同一特征區(qū)內(nèi),也會(huì)有各種因素對(duì)生物類(lèi)似性的確證產(chǎn)生不同程度影響。為在生物類(lèi)似藥物的特征區(qū)內(nèi)提供類(lèi)似性的證據(jù),本研究采用了包括主成分分析(PCA)在內(nèi)的不同加權(quán)分析法進(jìn)行了類(lèi)似性的試驗(yàn)分析和比較。并在2×2交叉設(shè)計(jì)基礎(chǔ)上進(jìn)行了廣泛的模擬研究,以對(duì)所提出的方法在有限樣本條件下的評(píng)價(jià)性能進(jìn)行研究。由于在特征區(qū)域內(nèi)也存在不同變量間的相關(guān)性,權(quán)重主成分分析(PCAweight)評(píng)價(jià)的把握度最高。該方法可進(jìn)一步拓展應(yīng)用于生物類(lèi)似藥物各種特征區(qū)域間的生物類(lèi)似性的"證據(jù)鏈完備性"評(píng)價(jià)。
[Abstract]:In order to evaluate the biosimilarity of bionic biological products (follow-on biologics), the (FDA) issued three draft guidelines in February 2012. In the draft guiding principles for "Scientific considerations" (scientific consideration), FDA requires that the similarity of products must be "totality-of-the-evidence" in each characteristic region, such as safety, purity and effectiveness. However, even in the same characteristic region, there are various factors which have different influence on the confirmation of biological similarity. In order to provide evidence of similarity in the characteristic region of biological analogues, different weighted analysis methods including principal component analysis (PCA) were used for experimental analysis and comparison of similarity. On the basis of 2 脳 2 cross design, a wide range of simulation studies are carried out in order to study the evaluation performance of the proposed method under the condition of limited samples. Because of the correlation between different variables in the characteristic region, the control degree of weight principal component analysis (PCAweight) is the highest. This method can be further applied to the "evidence chain completeness" evaluation of biological similarity among different characteristic regions of biological analogues.
