右美托咪定對(duì)小鼠視網(wǎng)膜缺血再灌注損傷的保護(hù)機(jī)制

【摘要】:目的研究右美托咪定對(duì)小鼠視網(wǎng)膜缺血再灌注損傷的保護(hù)作用機(jī)制。方法選取8周齡C57BL/6雄性小鼠48只,按照體重隨機(jī)分為3組:假手術(shù)組、模型組和實(shí)驗(yàn)組,每組16只。制備小鼠視網(wǎng)膜缺血再灌注損傷(RIRI)模型。在缺血前15 min和再灌注前5 min,實(shí)驗(yàn)組小鼠分別腹腔注射右美托咪定25μg·kg~(-1),假手術(shù)組和模型組腹腔注射同等劑量的0.9%Na Cl。在模型建立成功后,于再灌注24 h處死并取材。取小鼠視網(wǎng)膜組織勻漿液,用四唑鹽試劑(WST-1)法檢測(cè)超氧化物歧化酶(SOD),用TBA法檢測(cè)丙二醛(MDA),用比色法測(cè)定谷胱甘肽過(guò)氧化物酶(GSH-PX),用酶聯(lián)免疫吸附法檢測(cè)腫瘤壞死因子α(TNF-α)、白細(xì)胞介素-6(IL-6)、1-甲基環(huán)丙烯(1-MCP)及IL-10水平。結(jié)果與假手術(shù)組比較,模型組小鼠視網(wǎng)膜組織中SOD為(45.47±8.16)U·mg~(-1)、GSH-PX為(264.64±27.31)U·mg~(-1),均明顯降低;而MDA為(1.56±0.41)nmol·mg~(-1)、TNF-α為(2.67±0.23)ng·mL~(-1)、IL-6為(2.84±0.34)ng·mL~(-1)、MCP-1為(0.68±0.06)ng·mL~(-1)和IL-10為(0.21±0.02)ng·mL~(-1),均明顯升高,差異有統(tǒng)計(jì)學(xué)意義(均P0.05)。與模型組比較,實(shí)驗(yàn)組小鼠視網(wǎng)膜組織中SOD為(71.05±9.34)U·mg~(-1)、GSH-PX為(382.20±31.56)U·mg~(-1)、IL-10為(0.44±0.07)ng·mL~(-1),均明顯降低;而MDA為(1.02±0.23)nmol·mg~(-1)、TNF-α為(1.53±0.20)ng·mL~(-1)、IL-6為(1.64±0.07)ng·mL~(-1)、MCP-1為(0.41±0.07)ng·mL~(-1),均明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論右美托咪定可以改善視網(wǎng)膜缺血再灌注損傷,其機(jī)制與抑制氧自由基所致的脂質(zhì)過(guò)氧化損傷和抑制炎癥因子的分泌有關(guān)。
[Abstract]:Objective to study the protective effect of right metametrimidine on retinal ischemia reperfusion injury in mice. Methods 48 8-week-old male C57BL/6 mice were randomly divided into three groups according to their weight: sham-operated group, model group and experimental group, with 16 mice in each group. The (RIRI) model of retinal ischemia reperfusion injury in mice was established. At 15 min before ischemia and 5 min, before reperfusion, the mice in the experimental group were intraperitoneally injected with 25 渭 g kg~ (- 1) of right metomidine, and the same dose of 0.9%Na Cl. was injected intraperitoneally in the sham-operated group and the model group. After the successful establishment of the model, the rats were killed at 24 h after reperfusion and the samples were obtained. The contents of superoxide dismutase (SOD), malondialdehyde (MDA),) and glutathione peroxidase (GSH-PX) in retinal homogenate of mice were determined by WST-1 method and TBA method, and glutathione peroxidase (GSH-PX) was determined by colorimetric method. The levels of tumor necrosis factor-偽 (TNF- 偽), interleukin-6 (IL-6), 1-methylcyclopropene (1-MCP) and IL-10 were measured by enzyme-linked immunosorbent assay (Elisa). Results compared with the sham-operated group, the SOD and GSH-PX in the retinal tissue of the model group were (45.47 鹵8.16) U mg~ (- 1) and (264.64 鹵27.31) U mg~ (- 1). While MDA, TNF- 偽 and IL-6 were (1.56 鹵0.41) nmol mg~ (- 1), (2.67 鹵0.23) ng 路mL ~ (- 1) and (2.84 鹵0.34) ng 路mL ~ (- 1), respectively. MCP-1 was (0.68 鹵0.06) ng 路mL ~ (- 1) and IL-10 was (0.21 鹵0.02) ng 路mL ~ (- 1), the difference was statistically significant (P 0.05). Compared with the model group, the SOD, GSH-PX and IL-10 were (71.05 鹵9.34) U mg~ (- 1), (382.20 鹵31.56) U mg~ (- 1) and (0.44 鹵0.07) ng 路mL ~ (- 1) in the experimental group. All of them decreased significantly. While MDA, TNF- 偽, IL-6 and MCP-1 were (1.02 鹵0.23) nmol mg~ (- 1), (1.53 鹵0.20) ng 路mL ~ (- 1), (1.64 鹵0.07) ng 路mL ~ (- 1) and (0.41 鹵0.07) ng 路mL ~ (- 1), respectively. The difference was statistically significant (P0.05). Conclusion right metametrimidine can improve retinal ischemia-reperfusion injury by inhibiting lipid peroxidation injury induced by oxygen free radicals and inhibiting the secretion of inflammatory factors.
