納米氧化銅對豚鼠血腦屏障通透性影響觀察
[Abstract]:Objective to observe the effect of 40 and 200 nm nano-sized copper oxide particles (Cu ONPs) on the permeability of blood-brain barrier in guinea pigs. Methods the near spherical Cu ONPs with particle size of 40 nm and 200 nm was prepared for testing. The dispersion suspension of Cu ONPs was prepared by using 0.5%HPMC as dispersant. A guinea pig of 250 g 300 g was selected for the experiment. Five animals in each group were divided into four groups: 40 and 200 nm Cu ONPs groups, Cu SO4 group, 0.5%HPMC group and saline group. . Cu ONPs group, Cu SO4 group, HPMC group and saline group were administrated according to 1.5 ml/ kg 路bw, 65 ml/ kg 路bw, 4 ml/ kg 路bw and 4 ml/ kg 路bw, respectively. Five animals in each group were taken from each group at 24 and 48 h after exposure. The animals were injected with 2% Evans blue (EB) saline solution (4 ml/kg) through the sublingual vein, and then cardiac perfusion was performed 1 hour later. The parietal lobe cortex and subcortical structure tissue were removed and the opening degree of blood-brain barrier (BBB) was indicated by the amount of EB infiltration in brain tissue (measured by absorbance A value). Results the A value of cortex and subcortical structure tissue exposed to 40 nm Cu ONP for 24 and 48 h was significantly higher than that of the corresponding 200 nm Cu ONP group and physiological saline group (P0.05). The A values of the two Cu ONPs groups treated for 48 h were significantly higher than those of the control group (P0.05). There was significant difference in A value between the two groups at 24 h (P0.05). At the same time, the cortical A value of 200 nm Cu ONPs group (0.383 鹵0.017 A/mg) was significantly different from that of physiological saline group (0.103 鹵0.021 A/mg) (P0.05). Except the A value of subcortical structure in Cu SO4 group was 0.087 鹵0.026 and 0.071 鹵0.031 A/mg higher than that in normal saline group (0.041 鹵0.013 and 0.069 鹵0.022 A/mg) at 24 and 48 hours, respectively. There was no significant difference between the other groups in A value and saline group (P0.05). Conclusion 40 nm Cu ONP has a significant open effect on blood-brain barrier in guinea pigs, and this effect can be enhanced at 24 h and 48 h.
