【摘要】：目的:探討鞘內(nèi)注射P2X7受體拮抗劑考馬斯亮藍(lán)(BBG)在福爾馬林模型大鼠中的鎮(zhèn)痛作用及對(duì)脊髓磷脂酶A2(PLA2)表達(dá)的影響。方法:雄性Wistar大鼠鞘內(nèi)置管成功后鞘內(nèi)注射P2X7受體拮抗劑考馬斯亮藍(lán)(BBG)后所有的大鼠左側(cè)后肢爪掌背側(cè)皮下注射福爾馬林(5%,50μl),觀察大鼠在第一相(0~10 min)和第二相(10~60 min)的疼痛行為學(xué)比,變化(自發(fā)縮足次數(shù))。免疫組織化學(xué)方法檢測脊髓背角PLA2蛋白的表達(dá)與變化。結(jié)果:各組間大鼠第一相相比自發(fā)縮足次數(shù)沒有明顯的差異;第二相與對(duì)照組相比鞘內(nèi)1μg、5μg及腹腔10μg BBG注射組相比,鞘內(nèi)10μg BBG注射組大鼠的疼痛行為學(xué)明顯減少(自發(fā)縮足次數(shù),P0.05)。與此同時(shí),鞘內(nèi)10μg BBG注射組大鼠脊髓背角淺層內(nèi)PLA_2免疫陽性產(chǎn)物的表達(dá)與其它組比較明顯減少(P0.05)。結(jié)論:鞘內(nèi)注射10μg BBG可減輕福爾馬林誘導(dǎo)的炎性痛覺過敏,可能是通過抑制脊髓背角淺層PLA2的表達(dá)而發(fā)揮作用。
[Abstract]:Aim: to investigate the analgesic effect of intrathecal injection of P2X7 receptor antagonist Coomassie brilliant blue (BBG) in formalin model rats and the effect of intrathecal injection on the expression of phospholipase A 2 (PLA2) in spinal cord. Methods: after intrathecal injection of P2X7 receptor antagonist Coomassie brilliant blue (BBG), formalin (5%, 50 渭 l) was subcutaneously injected into the dorsal claw metacarpal of the left hind limb of male Wistar rats. The pain behavior ratios and changes in the first phase (0? 10 min) and the second phase (10? 60 min) were observed. Immunohistochemical method was used to detect the expression and change of PLA2 protein in spinal dorsal horn. Results: there was no significant difference in the frequency of spontaneous foot contraction between the first phase and the first phase of the rats. In the second phase, compared with the control group, 1 渭 g, 5 渭 g intrathecal and 10 渭 g BBG injected intrathecally, the pain behavior of the rats in the intrathecal 10 渭 g BBG injection group was significantly decreased (P 0.05). At the same time, the expression of PLA_2 immunopositive products in the superficial layer of spinal dorsal horn in the intrathecal 10 渭 g BBG injection group was significantly lower than that in the other groups (P0.05). Conclusion: intrathecal injection of 10 渭 g BBG can reduce formalin-induced inflammatory hyperalgesia, which may be due to the inhibition of the expression of PLA2 in the spinal dorsal horn.
