孤兒藥研發(fā)相關(guān)的信息學(xué)研究進(jìn)展

【摘要】:藥物研發(fā)包括臨床前研究和臨床研究,在推動(dòng)醫(yī)藥產(chǎn)業(yè)發(fā)展中發(fā)揮著至關(guān)重要的作用。由于研發(fā)各階段投入成本高、周期長,藥物毒性作用難預(yù)知等原因,新藥開發(fā)的耗費(fèi)巨大,成功率平均只有萬分之一,周期可長達(dá)幾年甚至幾十年。而罕見病藥物研發(fā)由于基因異質(zhì)性高、病理機(jī)制未明、可參與臨床試驗(yàn)的患者數(shù)量少等問題在藥物研發(fā)上更是具有較大局限性。近年來信息學(xué)領(lǐng)域的飛速發(fā)展大大推動(dòng)了藥物研發(fā)的進(jìn)程,基于計(jì)算機(jī)的理性藥物設(shè)計(jì)、海量虛擬篩選、組合化學(xué)、高通量反應(yīng)篩選系統(tǒng)等技術(shù)不斷發(fā)展成熟,在新藥研發(fā)的各個(gè)階段均起到積極的促進(jìn)作用;信息技術(shù)還有助于藥物重定位這一新型藥物開發(fā)模式的發(fā)展,通過藥物警戒作用可對(duì)藥物研發(fā)進(jìn)行有效的反饋、補(bǔ)充。信息技術(shù)的發(fā)展還促進(jìn)了藥物研發(fā)模式向集成式研發(fā)轉(zhuǎn)變。這對(duì)孤兒藥研發(fā)起到了積極的促進(jìn)作用。本文主要對(duì)孤兒藥研發(fā)相關(guān)的信息學(xué)研究進(jìn)展進(jìn)行綜述。
[Abstract]:Drug research and development, including pre-clinical research and clinical research, plays an important role in promoting the development of pharmaceutical industry. Due to the high cost of R & D, long cycle and difficult to predict the toxicity of drugs, the cost of new drug development is huge, the average success rate is only 1/10000, and the cycle can be as long as several years or even decades. However, drug development in rare diseases has great limitations due to its high gene heterogeneity, unclear pathological mechanism and the small number of patients who can participate in clinical trials. In recent years, the rapid development in the field of informatics has greatly promoted the process of drug research and development. Computer-based rational drug design, mass virtual screening, combinatorial chemistry, high-throughput reaction screening system and other technologies have been developed and matured. It has played a positive role in promoting the development of new drugs at every stage of the research and development of new drugs; Information technology also contributes to the development of a new drug development model, drug repositioning, which can provide effective feedback and supplement to drug development through pharmacovigilance. The development of information technology has also promoted the transformation of drug R & D model to integrated R & D. This has played a positive role in the development of orphan drugs. In this paper, the advances in informatics related to orphan drug development are reviewed.
