載RGD肽脂質(zhì)體的優(yōu)化和初步評價

【摘要】:采用逆相蒸發(fā)法制備了載RGD肽脂質(zhì)體。以包封率為指標(biāo),采用單因素試驗和正交設(shè)計優(yōu)化工藝條件。確定的優(yōu)化條件為:卵磷脂和膽固醇的質(zhì)量比為4∶1,磷酸鹽緩沖液的體積為10 ml,RGD肽的加入量為18 mg,超聲時間為3 min。3批優(yōu)化制品平均粒徑為473 nm,平均包封率為75.35%。RGD肽脂質(zhì)體在磷酸鹽緩沖液中16 h累積釋放率為48.5%,相較于RGD肽溶液有一定的緩釋效果。初步穩(wěn)定性試驗表明,載RGD肽脂質(zhì)體在4℃保存相對較穩(wěn)定。
[Abstract]:RGD peptide liposomes were prepared by reverse phase evaporation. The single factor test and orthogonal design were used to optimize the technological conditions with the inclusion efficiency as the index. The optimum conditions are as follows: the mass ratio of lecithin to cholesterol is 4 渭 1, the volume of phosphate buffer is 10 ml,RGD peptide, the dosage of peptide is 18 mg, the ultrasonic time is 3 min.3, the average particle size of the batch optimized product is 473 nm,. The average entrapment efficiency of 75.35%.RGD peptide liposomes was 48.5% in phosphate buffer solution for 16 h, which was more effective than that of RGD peptide solution. The preliminary stability test showed that the RGD-loaded peptide liposomes were relatively stable at 4 鈩,
