【摘要】：目的建立LC-MS/MS法檢測(cè)小鼠血漿內(nèi)FG-4592的濃度,研究FG-4592在小鼠體內(nèi)的藥代動(dòng)力學(xué)。方法KM小鼠腹腔注射FG-4592 5 mg/kg,不同時(shí)間點(diǎn)采集血漿,樣品經(jīng)甲醇沉淀。選用Agilent Zorbax SB-Aq-C18色譜柱(2.1 mm×100 mm,3.5μm),柱溫25℃,流動(dòng)相為甲醇-水(含5 mmol/L甲酸銨,0.1%甲酸)(90∶10),流速0.35 m L/min。采用ESI離子源,正離子模式多反應(yīng)監(jiān)測(cè),離子通道分別為m/z 353.1→m/z 278.1(FG-4592)和m/z294.9→m/z 235.0(內(nèi)標(biāo)物)。結(jié)果測(cè)定血漿中FG-4592的線性范圍為1~1000 ng/m L,日內(nèi)及日間精密度均10%,準(zhǔn)確度在90%~105%范圍內(nèi),方法學(xué)考察均符合生物樣品的分析要求。藥代動(dòng)力學(xué)參數(shù):Cmax為(22.80±0.36)μg/m L,AUC(0-72)為(31.64±0.97)(μg·h)/m L,t1/2為8.81 h。結(jié)論本研究建立的FG-4592血藥濃度測(cè)定方法可行,該藥物在血液中吸收良好,測(cè)定結(jié)果可為藥物的基礎(chǔ)研究提供依據(jù)。
