依折麥布及其R-對(duì)映體的超臨界流體色譜手性拆分方法研究

【摘要】:目的建立膽固醇抑制吸收劑依折麥布及其R-對(duì)映體的超臨界流體色譜手性拆分方法。方法采用Chiralcel OD色譜柱(4.6 mm×250 mm,10μm);以超臨界二氧化碳-含0.1%三氟乙酸-0.1%三乙胺的甲醇(90∶10)為流動(dòng)相,背壓為15MPa,進(jìn)樣量為5μL,柱溫為35℃,流速為3.0 m L·min-1,于235 nm波長(zhǎng)處分離依折麥布及其對(duì)映體。結(jié)果依折麥布與R-對(duì)映體在15 min內(nèi)依次出峰,分離度為1.6;兩者分別在0.010~0.100 mg·m L-1內(nèi)線性關(guān)系良好,相關(guān)系數(shù)均為0.999 9(n=7)。依折麥布及其R-對(duì)映體的定量限分別為34、32 ng(S/N≥10),檢測(cè)限分別為10、9 ng(S/N≥3)。原料中R-對(duì)映體的平均加樣回收率為98.4%(n=9)。結(jié)論超臨界流體色譜作為一種綠色環(huán)保的色譜方法,分離效率高,重現(xiàn)性好,可有效用于依折麥布原料及制劑中R-對(duì)映體的測(cè)定及質(zhì)量控制。
