以ANO1蛋白為靶點(diǎn)的鈣激活氯通道調(diào)節(jié)劑研究進(jìn)展

【摘要】:鈣激活氯通道是廣泛存在于人體內(nèi)的一種陰離子通道,參與分泌、心肌細(xì)胞去極化、神經(jīng)信號(hào)傳導(dǎo)等多種生理活動(dòng)。ANO1蛋白是鈣激活氯通道的分子基礎(chǔ)之一,對(duì)ANO1蛋白進(jìn)行調(diào)節(jié)能夠產(chǎn)生多種藥效作用,例如鎮(zhèn)痛、治療痢疾、哮喘、抑制腫瘤等。近10年來(lái)發(fā)展出許多ANO1蛋白調(diào)節(jié)劑篩選測(cè)試方法,雖然篩選出一系列以ANO1蛋白為靶點(diǎn)的鈣激活氯通道調(diào)節(jié)分子,但是這些分子的藥理學(xué)作用卻并不一致。本文從篩選方法、構(gòu)效關(guān)系、潛在應(yīng)用前景等多方面對(duì)ANO1蛋白調(diào)節(jié)分子的研究進(jìn)展進(jìn)行論述。
[Abstract]:Calcium-activated chloride channel is a kind of anionic channel which is widely present in human body. It is involved in many physiological activities such as secretion, depolarization of cardiac myocytes, nerve signal transduction and so on. ANO1 protein is one of the molecular bases of calcium-activated chloride channel. Regulation of ANO1 protein can produce a variety of pharmacological effects, such as analgesia, dysentery, asthma, tumor suppression and so on. In recent 10 years, many screening and testing methods for ANO1 protein modulators have been developed. Although a series of calcium-activated chloride channel regulators targeting ANO1 proteins have been screened, the pharmacological effects of these molecules are not consistent. In this paper, the research progress of regulatory molecules of ANO1 proteins is reviewed in terms of screening methods, structure-activity relationships and potential application prospects.
