新疆馬脂不飽和脂肪酸軟膠囊的制備工藝研究

【摘要】:目的:采用壓榨法及溶劑結(jié)晶法對馬脂不飽和脂肪酸進(jìn)行富集提取,其主要功效成分為油酸及亞油酸,并篩選添加抗氧化劑,研制馬脂不飽和脂肪酸軟膠囊。方法:1.以酸值、碘值、過氧化值及產(chǎn)率為考察指標(biāo),采用單因素試驗(yàn),優(yōu)選馬油毛油的提取工藝。2.采用溶劑結(jié)晶法對馬脂不飽和脂肪酸進(jìn)行提取,對所得油脂氣相色譜質(zhì)譜聯(lián)用技術(shù)分析馬脂脂肪酸成分,以油酸綜合產(chǎn)量為考察指標(biāo),通過單因素試驗(yàn)篩選最佳溶劑種類及最佳提取次數(shù)并采用正交試驗(yàn)考察結(jié)晶溫度、結(jié)晶時間及混合脂肪酸與溶劑比例對馬脂不飽和脂肪酸溶劑結(jié)晶的影響。3.以馬脂不飽和脂肪酸作為軟膠囊主成分,采用單因素考察法篩選抗氧化劑種類及添加量,確定軟膠囊制備工藝。結(jié)果:1.壓榨法影響因素中不同提取時間的影響比不同提取溫度的影響更大;最佳提取工藝為:提取時間6h,提取溫度80-C;此條件下數(shù)據(jù)結(jié)果最為理想,產(chǎn)率為39.44±0.07g,酸值碘值過氧化值分別為3.580mg/g,81.012g/100g,0.916mmol/kg。2.溶劑結(jié)晶提取工藝:通過單因素試驗(yàn)得到:最佳溶劑是95%乙醇,最佳工藝條件是結(jié)晶溫度2。C,最佳結(jié)晶時間6h,最佳混合脂肪酸與溶劑比例是1:7;濾餅進(jìn)行重復(fù)結(jié)晶的最佳次數(shù)為兩次;通過正交試驗(yàn)得到最佳的工藝條件是:混合脂肪酸與溶劑比例是1:9;結(jié)晶時間為8h;結(jié)晶溫度為2。C。3.添加抗氧化劑的最佳種類為維生素E,最佳添加量為0.02%,軟膠囊囊殼中的成分明膠甘油水的最佳比例為1:0.5:1。結(jié)論:壓榨法相對溶劑提取法更方便、安全,且產(chǎn)率較高,適合大規(guī)模工業(yè)生產(chǎn);溶劑結(jié)晶法提取后不飽和脂肪酸含量較提取前提高了24.39%,油酸含量提高了20.21%。經(jīng)溶劑結(jié)晶法提取后的馬油,含有更高的單不飽和脂肪酸,可開發(fā)為極具潛力的保健品。
[Abstract]:Objective: to enrich and extract the unsaturated fatty acids of equine by squeezing and solvent crystallization. The main components are oleic acid and linoleic acid. The soft capsules of unsaturated fatty acids of equine fat were prepared by screening and adding antioxidants. Methods: 1. With acid value, iodine value, peroxide value and yield as the index of investigation, single factor test was used to optimize the extraction process of horse oil wool oil. 2. The unsaturated fatty acids of horse fat were extracted by solvent crystallization method. The fatty acids were analyzed by GC-MS. The comprehensive yield of oleic acid was used as the index. The effects of crystallization temperature, crystallization time and the ratio of mixed fatty acids to solvent on the solvent crystallization of unsaturated fatty acids were investigated by orthogonal test. Taking the unsaturated fatty acid of horse fat as the main component of soft capsule, the kinds of antioxidants and the amount of antioxidants were selected by single factor investigation method, and the preparation process of soft capsule was determined. Results: 1. The influence of different extraction time was greater than that of different extraction temperature, and the optimum extraction technology was as follows: extraction time 6 h, extraction temperature 80-C; Under this condition, the yield is 39.44 鹵0.07g, the peroxide value of acid value iodine is 3.580mg / g / 100g / 100mg / 100mg 路kg ~ (-2), and the value of peroxide value is 3.580mg / g / 100g / 100g / 100g / 100g ~ (-1) 路g ~ (-1), respectively. The solvent crystallization extraction process: the optimum solvent is 95% ethanol, the optimum condition is crystallization temperature 2.C, the optimum crystallization time is 6 h, the optimum ratio of mixed fatty acid to solvent is 1: 7; The optimum process conditions are as follows: the ratio of mixed fatty acid to solvent is 1: 9, the crystallization time is 8 h, and the crystallization temperature is 2.C. 3, the optimum times of repeated crystallization of filter cake are two times, and the optimum technological conditions are as follows: the ratio of mixed fatty acid to solvent is 1: 9; the crystallization time is 8 h; The best type of antioxidants was vitamin E, the optimum addition amount was 0.02, and the optimum proportion of gelatin glycerin water in capsule shell was 1: 0.5: 1. Conclusion: the squeezing method is more convenient, safe and suitable for large-scale industrial production than solvent extraction, and the content of unsaturated fatty acids and oleic acid after solvent crystallization is 24.39 and 20.21 respectively. The horse oil extracted by solvent crystallization method contains higher monounsaturated fatty acids and can be developed as a potential health product.
