【摘要】：目的研究抑制囊泡谷氨酸轉(zhuǎn)運(yùn)體(vesicular glutamate transporters,VGLUT)功能對(duì)動(dòng)物疼痛行為的影響。方法分別觀察選擇性VGLUT抑制劑芝加哥天藍(lán)6B(Chicago sky blue 6B,CSB6B)對(duì)小鼠熱板潛伏期、醋酸扭體次數(shù)和福爾馬林所致舔足時(shí)間的影響。結(jié)果腹腔注射CSB6B不影響小鼠熱板潛伏期。小鼠醋酸扭體實(shí)驗(yàn)中,與溶劑對(duì)照組相比,CSB6B 2.5mg/kg腹腔注射30 min后可使小鼠扭體次數(shù)從26.50±2.97顯著下降到8.22±1.90(P0.01),腹腔注射60 min后可使小鼠扭體次數(shù)顯著下降到9.60±1.84(P0.01)。在福爾馬林實(shí)驗(yàn)中,與溶劑對(duì)照組相比,CSB6B不影響小鼠的Ⅰ相舔足時(shí)間,CSB6B 0.5 mg/kg腹腔注射30 min后可使小鼠的Ⅱ相舔足時(shí)間從(139.40±21.02)s顯著下降到(75.10±19.45)s(P0.05),而注射2 h后對(duì)小鼠舔足時(shí)間沒(méi)有顯著影響。結(jié)論抑制VGLUT功能可顯著抑制炎性疼痛,提示VGLUT參與了炎性疼痛的調(diào)控,可能成為疼痛治療的新靶點(diǎn)。
[Abstract]:Objective to study the effects of inhibition of vesicular glutamate transporter (vesicular glutamate transporters,VGLUT) on pain behavior in animals. Methods the effects of selective VGLUT inhibitor B B (Chicago sky blue 6 B 6 B Results Intraperitoneal injection of CSB6B did not affect the latency of hot plate in mice. In the acetic acid writhing test, compared with the solvent control group, CSB6B 2.5mg/kg could significantly decrease the writhing times from 26.50 鹵2.97 to 8.22 鹵1.90 (P0.01) after 30 min intraperitoneal injection. After 60 min intraperitoneal injection, the number of writhing decreased to 9.60 鹵1.84 (P0.01). In the formalin experiment, compared with the solvent control group, CSB6B did not affect the lapping time of phase I of mice.
