樟芝酵素對苯丙胺致PC12細(xì)胞毒性損傷的保護(hù)作用

【摘要】:目的探討樟芝酵素對苯丙胺致PC12細(xì)胞損傷的保護(hù)作用及其機(jī)理。方法利用PC12細(xì)胞模型,設(shè)置對照組,3.5 mmol/L苯丙胺組和3.5 mmol/L苯丙胺+樟芝酵素組。(1)通過顯微鏡觀察細(xì)胞形態(tài)的變化;(2)通過MTT法檢測細(xì)胞存活率;(3)通過Western blot分析Caspase-3的變化;(4)通過流式細(xì)胞儀檢測細(xì)胞凋亡和細(xì)胞內(nèi)活性氧的變化。結(jié)果 (1)苯丙胺損傷了PC12細(xì)胞的結(jié)構(gòu),導(dǎo)致細(xì)胞凋亡增加,樟芝酵素能夠減少細(xì)胞的凋亡,提高細(xì)胞的存活率;(2)樟芝酵素減少了細(xì)胞內(nèi)ROS的生成;(3)樟芝酵素組procaspase-3(35 kd)蛋白含量比苯丙胺組增多,激活分子caspase-3(17 kd)含量較苯丙胺組減少。結(jié)論樟芝酵素對苯丙胺致PC12細(xì)胞毒性損傷具有保護(hù)作用,其機(jī)理與抑制細(xì)胞線粒體凋亡信號通路以及減少細(xì)胞內(nèi)氧自由基的產(chǎn)生密切相關(guān)。
[Abstract]:Objective to investigate the protective effect of camphor luciferase on PC12 cells induced by amphetamine and its mechanism. Methods the PC12 cell model was used to set up the control group, 3.5 mmol/L amphetamine group and 3.5 mmol/L phenylaniline camphor enzyme group. (1) the morphological changes of the cells were observed by microscope; (2) the cell survival rate was detected by MTT method. (3) the changes of Caspase-3 were analyzed by Western blot, and (4) the changes of apoptosis and intracellular reactive oxygen species were detected by flow cytometry. Results (1) the structure of PC12 cells was damaged by amphetamine, and the apoptosis was increased, and the cell apoptosis was decreased and the survival rate was improved by camphor enzyme, and (2) the production of ROS was decreased by anilaniline. (3) the protein content of procaspase-3 (35 kd) in anisodycin group was higher than that in amphetamine group, and the content of activator caspase-3 (17 kd) was lower than that in amphetamine group. Conclusion Cinnamomum camphora has protective effect on PC12 cell toxicity induced by amphetamine, and its mechanism is closely related to inhibition of mitochondrial apoptosis signaling pathway and reduction of intracellular oxygen free radical production.
