5-氟尿嘧啶聚乳酸緩釋片的處方篩選及體外釋藥機(jī)制研究

【摘要】:目的:篩選5-氟尿嘧啶(5-FU)聚乳酸(PLA)緩釋片(5-FU-PLA-DS)的處方,研究其體外釋藥機(jī)制。方法:采用紫外分光光度法測(cè)定釋放介質(zhì)中5-FU含量,以模擬體液為釋放介質(zhì),于37℃水浴中進(jìn)行體外釋放試驗(yàn)。以分子量分別為3 000、6 000、10 000、15 000和20 000的PLA,制備載藥量分別為1.5、2.5、3.0 mg/片,直徑3.0 mm、厚1.0 mm的15種圓形5-FU-PLA-DS;以有效濃度持續(xù)釋放時(shí)間與累積釋放度為指標(biāo),篩選最優(yōu)處方。采用掃描電鏡觀察5-FU-PLA-DS釋藥后的形態(tài),評(píng)價(jià)其釋藥機(jī)制。結(jié)果:最優(yōu)處方中PLA的分子量為20 000,載藥量為3.0 mg/片;所制5-FU-PLA-DS可釋放119 d,累積釋放度為100.63%,有效濃度持續(xù)釋放91 d。掃描電鏡觀察顯示,5-FU-PLA-DS表面的微球最先接觸釋放介質(zhì)而進(jìn)行蝕解、釋放藥物,待外層的微球和微球之間的PLA降解后才逐漸暴露出內(nèi)層的微球進(jìn)行釋藥,其釋藥機(jī)制主要為溶蝕和擴(kuò)散。結(jié)論:成功制得5-FU-PLA-DS,其有效濃度持續(xù)釋放時(shí)間長(zhǎng),能夠由外向內(nèi)有步驟地降解,實(shí)現(xiàn)不同層次微球的非同步釋藥。
