【摘要】：目的探討五味子乙素與脂質(zhì)體對(duì)克服腫瘤多藥耐藥的協(xié)同作用。方法制備獲得多柔比星-五味子乙素共載脂質(zhì)體,以人白血細(xì)胞系K562/DOX耐藥細(xì)胞為模型細(xì)胞,以多柔比星溶液、五味子乙素+多柔比星混合液、多柔比星脂質(zhì)體、多柔比星脂質(zhì)體與五味子乙素混合液、維拉帕米+多柔比星混合液為對(duì)照,MTT法測定細(xì)胞毒性,并測定不同時(shí)間點(diǎn)進(jìn)入耐藥細(xì)胞的多柔比星量,繪制藥物濃度隨時(shí)間變化曲線。結(jié)果多柔比星-五味子乙素共載脂質(zhì)體逆轉(zhuǎn)倍數(shù)分別是五味子乙素、脂質(zhì)體、多柔比星脂質(zhì)體與五味子乙素混合液的1.68、14.52和1.42倍。共載脂質(zhì)體使多柔比星進(jìn)入耐藥細(xì)胞的藥物量是多柔比星脂質(zhì)體、五味子乙素的1.30和1.21倍,且具有延緩藥物外排作用。結(jié)論多柔比星-五味子乙素共載脂質(zhì)體具備良好的克服腫瘤多藥耐藥作用,脂質(zhì)體和五味子乙素顯示了良好的協(xié)同作用。
[Abstract]:Objective to investigate the synergistic effect of Schisandrin and liposome on overcoming multidrug resistance of tumor. Methods doxorubicin and Schisandrin B co-loaded liposomes were prepared. Human white blood cell line K562/DOX resistant cells were used as model cells, doxorubicin solution, schisandrin doxorubicin mixture and doxorubicin liposome were prepared. Doxorubicin liposome, schisandrin B and verapamide-doxorubicin were used as control. The cytotoxicity was determined by MTT method, and the amount of doxorubicin entered into drug-resistant cells at different time points, and the change curve of drug concentration with time was plotted. Results the reversal times of doxorubicin and schisandrin B co-loaded liposomes were 1.68- 14.52 and 1.42 times of those of Schisandrin B liposome, liposomes, doxorubicin liposomes and schisandrin B liposomes respectively. Cocarrier liposomes could induce doxorubicin into drug-resistant cells by 1.30 and 1.21 times as much as doxorubicin liposomes and by 1.30 and 1.21 times as much as Schisandrin, and could delay drug efflux. Conclusion doxorubicin-Schisandrin-B cocarrier liposome has a good effect on overcoming multidrug resistance of tumor. Liposome and Schisandrin B have good synergistic effect.
