血液和尿液中吲哚美辛和布洛芬含量測(cè)定的方法學(xué)研究及其應(yīng)用

【摘要】:目的:非甾體抗炎藥(Nonsteroidal Antiinflammatory Drugs,NSAIDs)因?yàn)橛薪鉄、降低疼痛、?duì)抗炎癥和對(duì)抗風(fēng)濕等臨床作用而被廣泛的應(yīng)用于臨床。本實(shí)驗(yàn)擬建立一種同時(shí)測(cè)定血漿和尿液中吲哚美辛和布洛芬的藥物濃度的HPLC方法。并運(yùn)用該方法對(duì)實(shí)際樣品進(jìn)行測(cè)定,考察兩種藥物的血藥濃度變化情況。方法:ZORBAX SB-C18柱(4.6mm×250mm,5μm)色譜柱,流動(dòng)相為乙腈和醋酸銨溶液(20mmol/L,pH=3.5)=85:15(v/v),流速為1mL/min,進(jìn)樣量80μL,布洛芬保留時(shí)間為8.57min左右,吲哚美辛的保留時(shí)間6.78min左右,柱溫為室溫,檢測(cè)波長(zhǎng)220nm。結(jié)果:吲哚美辛和布洛芬能夠與內(nèi)源性物質(zhì)有效分離,在較寬的濃度范圍內(nèi)吲哚美辛和布洛芬的峰面積和與其濃度呈較好的線性關(guān)系(R"g0.9985),方法的精密度小于4.0%,方法回收率大于85%。生物樣品在4℃冰箱中放置48小時(shí)性質(zhì)穩(wěn)定。結(jié)論:本方法專屬性好,實(shí)驗(yàn)所用時(shí)間短?勺鳛樯飿悠分羞胚崦佬梁筒悸宸业乃幬餄舛葴y(cè)定的方法。運(yùn)用此方法對(duì)數(shù)只大鼠血藥濃度監(jiān)測(cè)結(jié)果表明,布洛芬在1h左右達(dá)到最大血藥濃度(27.65μg/mL),吲哚美辛在3h左右達(dá)最大血藥濃度(2.71μg/mL)。本研究為進(jìn)一步研究吲哚美辛和布洛芬體內(nèi)利用度奠定基礎(chǔ),為臨床安全合理用藥提供了依據(jù)。也為其他非甾體抗炎藥的分析提供了參考。
[Abstract]:Objective: Non-steroidal anti-inflammatory drug (Nonsteroidal Antiinflammatory Drugs,NSAIDs) has been widely used in clinic because of its antipyretic, pain reduction, anti-inflammatory and anti-rheumatic effects. A HPLC method for simultaneous determination of indomethacin and ibuprofen in plasma and urine was developed. The actual samples were determined by this method, and the changes of blood concentration of the two drugs were investigated. Methods: ZORBAX SB-C18 column (4.6mm 脳 250mm ~ 5 渭 m) was used. The mobile phase consisted of acetonitrile and ammonium acetate solution (20 mmol / L ~ (-1) pH ~ (-1) = 85:15 (v / v), the flow rate was 1 mL / min, the injection amount was 80 渭 L, and the retention time of ibuprofen was about 8.57min. The retention time of indomethacin was about 6.78min, the column temperature was room temperature and the detection wavelength was 220 nm. Results: indomethacin and ibuprofen could be effectively separated from endogenous substances. The peak area and concentration of indomethacin and ibuprofen were linear in a wide range of concentrations (R "g 0.9985).
