HPLC法測(cè)定兔眼角膜中艾沙康唑的濃度

【摘要】:目的:建立測(cè)定兔眼角膜中艾沙康唑濃度的方法。方法:采用高效液相色譜(HPLC)法。色譜柱為Waters X-Bridge C_(18),流動(dòng)相為0.2%冰乙酸-乙腈(49:51,V/V),流速為0.7 mL/min,柱溫為40℃,檢測(cè)波長(zhǎng)為284 nm,進(jìn)樣量為10μL。取日本大耳白兔15只,雙眼分別滴加0.2%艾沙康唑混懸液50μL,于給藥后5、15、30、60、90 min分別處死3只兔并剖取其角膜樣品,處理后進(jìn)樣測(cè)定艾沙康唑濃度。結(jié)果:角膜樣品中的艾沙康唑在0.031 2~2.24μg/mL質(zhì)量濃度范圍內(nèi)線性關(guān)系良好(r~2=0.999 9),方法回收率為91.7%~105.6%(RSD≤6.88%,n=5),提取回收率為100.50%~106.10%(RSD≤2.16%,n=3),日內(nèi)(n=5)、日間(n=3)精密度和穩(wěn)定性(n=5)試驗(yàn)的RSD均小于10%。結(jié)論:本方法專屬性強(qiáng)、準(zhǔn)確度高、重現(xiàn)性好,可快速、準(zhǔn)確地測(cè)定兔眼角膜中艾沙康唑的濃度。
