一氧化碳發(fā)色羅非魚片急性毒性與遺傳毒性研究

【摘要】:采用小鼠急性毒性實驗、鼠傷寒沙門氏菌回復(fù)突變實驗(Ames)、小鼠骨髓細胞微核實驗、小鼠睪丸染色體畸變實驗研究發(fā)色羅非魚片的急性毒性和遺傳毒性。結(jié)果表明,小鼠按照急性經(jīng)口最大耐受劑量26.4 g/kg體重給予受試物后,14 d內(nèi)觀察實驗雌性和雄性小鼠均未出現(xiàn)死亡情況。遺傳毒性實驗表明,發(fā)色羅非魚片50、500、5000μg的處理組TA1535、TA1537、TA1538、TA98、TA100的鼠傷寒沙門氏菌回變菌落數(shù)最大值為(165±7)個/皿,顯著低于添加二硝酸氟(NF)和2-苯蒽胺(ANTH)的陽性對照組(p0.01);高、中、低劑量組小鼠骨髓微核率分別為(1.0‰±0.5‰)、(0.3‰±0.5‰)和(0.5‰±0.7‰),劑量組之間無明顯的劑量關(guān)系;高、中、低劑量組睪丸染色體總畸變率分別為5.5%、5.9%和4.9%,與陰性對照組總畸變率4.8%相比無顯著性差異,明顯低于陽性組(p0.01)。說明本次實驗條件下,發(fā)色羅非魚片經(jīng)口最大耐受劑量(MTD)15 g/kg安全劑量,Ames實驗、小鼠骨髓微核實驗和睪丸染色體畸變實驗均呈陰性,說明CO發(fā)色羅非魚片無急性毒性及遺傳毒性。
[Abstract]:The acute toxicity and genetic toxicity of chromatic tilapia tablets were studied by mouse acute toxicity test, (Ames), mouse bone marrow micronucleus test and mouse testicular chromosome aberration test. The results showed that there was no death in both female and male mice within 14 days after the mice were given acute oral maximum tolerance dose of 26.4 g/kg. The genotoxicity test showed that the maximum number of reversion colonies of Salmonella typhimurium in TA1535,TA1537,TA1538,TA98,TA100 treated group was (165 鹵7) / dish. It was significantly lower than that of the positive control group supplemented with fluorine dinitrate (NF) and 2-phenanthlamine (ANTH) (p0.01).
