【摘要】：青霉素結(jié)合蛋白(PBPs)是一類廣泛存在于細(xì)菌細(xì)胞膜表面的膜蛋白,是β-內(nèi)酰胺類抗生素的主要作用靶位。在細(xì)菌合成細(xì)胞壁肽聚糖的過程中,PBPs主要發(fā)揮糖基轉(zhuǎn)移酶、肽基轉(zhuǎn)移酶和D-丙氨酰-D-丙氨酸羧肽酶(D,D-羧肽酶)活性,是細(xì)菌生長(zhǎng)繁殖中不可或缺的酶。不同種類細(xì)菌所含PBPs各不相同,其結(jié)構(gòu)的改變、數(shù)量的增多、與抗生素親和力的下降以及產(chǎn)生新的青霉素結(jié)合蛋白是直接導(dǎo)致細(xì)菌對(duì)β-內(nèi)酰胺類抗生素產(chǎn)生耐藥性的重要原因。隨著各類抗菌藥物在臨床上的廣泛應(yīng)用,細(xì)菌對(duì)抗菌藥物的耐藥問題日趨嚴(yán)重,其耐藥水平也越來越高。因此,近年來全球圍繞PBPs開展的研究工作越來越多。本文對(duì)PBPs的分類、結(jié)構(gòu)和功能、與細(xì)菌耐藥性的關(guān)系及檢測(cè)方法的最新研究進(jìn)展進(jìn)行綜述,并對(duì)未來可能的研究方向進(jìn)行展望。
[Abstract]:Penicillin binding protein (PBPs) is a kind of membrane protein which widely exists on the surface of bacterial cell membrane and is the main target of 尾-lactam antibiotics. In the process of bacterial cell wall peptidoglycan synthesis, PBPs mainly plays an important role in bacterial growth and reproduction because of its activities of glycosyltransferase, peptidyl transferase and D-alanyl-D-alanine carboxypeptidase. Different species of bacteria contain different PBPs, the structure of the changes, the increase in the number, The decrease of affinity to antibiotics and the production of new penicillin binding proteins are the main causes of bacterial resistance to 尾-lactam antibiotics. With the wide application of various kinds of antimicrobial agents in clinic, the problem of bacterial resistance to antimicrobial agents is becoming more and more serious, and the level of drug resistance is becoming higher and higher. Therefore, in recent years, the global research work around PBPs is more and more. In this paper, the classification, structure and function of PBPs, the relationship between PBPs and bacterial resistance and the latest research progress of detection methods are reviewed, and the possible research directions in the future are prospected.
