地佐辛對(duì)坐骨神經(jīng)慢性壓迫性疼痛大鼠脊髓膠質(zhì)纖維酸性蛋白表達(dá)的影響

【摘要】:目的探索地佐辛對(duì)坐骨神經(jīng)慢性壓迫性疼痛(CCI)大鼠脊髓膠質(zhì)纖維酸性蛋白(GFAP)的影響及對(duì)坐骨神經(jīng)的保護(hù)作用。方法按照體重將SD大鼠隨機(jī)為4組,每組8只:假手術(shù)組、模型組和低、高2個(gè)劑量實(shí)驗(yàn)組(地佐辛,2.5,10 mg·kg~(-1))。于手術(shù)后即刻至術(shù)后7 d,各組每天腹腔注射藥物1次,假手術(shù)組和模型組給予等體積的0.9%NaCl。于給藥7 d后,電鏡下觀察各組坐骨神經(jīng)的病理形態(tài)學(xué)變化,以免疫熒光組織化學(xué)法觀測各組脊髓GFAP表達(dá)。結(jié)果地佐辛連續(xù)給藥7 d后,低劑量實(shí)驗(yàn)組大鼠的坐骨神經(jīng)纖維腫脹,鞘細(xì)胞數(shù)目減少,部分髓鞘溶解;高劑量實(shí)驗(yàn)組大鼠有少數(shù)正常髓鞘,較模型組神經(jīng)纖維腫脹減輕,但較假手術(shù)組鞘細(xì)胞減少,部分髓鞘中可見軸突。大鼠脊髓中GFAP的表達(dá)水平:假手術(shù)組、模型組、低、高2個(gè)劑量實(shí)驗(yàn)組分別為1.03±0.28,2.08±0.55,1.61±0.14,1.25±0.45。與假手術(shù)組比較,模型組差異有統(tǒng)計(jì)學(xué)意義(P0.001);與模型組比較,低、高2個(gè)劑量實(shí)驗(yàn)組差異均有統(tǒng)計(jì)學(xué)意義(P0.05,P0.01)。結(jié)論地佐辛對(duì)神經(jīng)病理性疼痛的抑制作用,可能與下調(diào)脊髓GFAP的表達(dá)、影響星形膠質(zhì)細(xì)胞的活化和減輕坐骨神經(jīng)水腫有密切關(guān)聯(lián)。
[Abstract]:Objective to investigate the effect of dizosin on glial fibrillary acidic protein (GFAP) (GFAP) and its protective effect on sciatic nerve in (CCI) rats with chronic compression pain of sciatic nerve. Methods according to body weight, SD rats were randomly divided into 4 groups with 8 rats in each group: sham-operated group, model group, low dose and high dose group (dizosin, 2.5 min 10 mg kg~ (-1). From immediately after operation to 7 days after operation, each group was intraperitoneally injected with drug once a day. The sham operation group and the model group were given 0.9 sodium chloride of the same volume. The histopathological changes of sciatic nerve in each group were observed under electron microscope and the expression of GFAP in spinal cord of each group was observed by immunofluorescence histochemical method 7 days after administration. Results after continuous administration of dizosin for 7 days, the sciatic nerve fibers in the low-dose experimental group were swollen, the number of sheath cells decreased, and some myelins were dissolved. There were a few normal myelin sheath in the high dose experimental group, which was less than that in the model group, but decreased compared with the sham operation group, and some axons were found in the myelin sheath. The expression level of GFAP in spinal cord of rats: sham operation group, model group, low dose and high dose group were 1.03 鹵0.28 鹵2.08 鹵0.55V 1.61 鹵0.141.61 鹵0.141.1.25 鹵0.45 respectively. Compared with sham-operation group, the difference between model group and model group was statistically significant (P0.001), compared with model group, the difference of low dose and high dose group was statistically significant (P0.05 + P0.01). Conclusion the inhibitory effect of dizosin on neuropathic pain may be related to the down-regulation of GFAP expression in spinal cord, the activation of astrocytes and the reduction of sciatic nerve edema.
