白藜蘆醇對甲基乙二醛損傷BRL-3A細(xì)胞的抑制作用

【摘要】:為研究甲基乙二醛(MGO)對大鼠肝細(xì)胞BRL-3A細(xì)胞的損傷機(jī)制,在MGO存在的情況下,以50μmol/L白藜蘆醇處理BRL-3A,分別用MTT法檢測細(xì)胞活力;DNPH法檢測細(xì)胞內(nèi)Schiff base濃度;DCFH-DA熒光探針法檢測ROS水平;以及脂質(zhì)過氧化產(chǎn)物MDA含量的檢測。結(jié)果表明,MGO導(dǎo)致細(xì)胞內(nèi)Schiff base、ROS和MDA顯著增加,誘導(dǎo)細(xì)胞發(fā)生氧化應(yīng)激,白藜蘆醇處理可有效改善MGO對細(xì)胞的損傷作用。
[Abstract]:In order to study the damage mechanism of methylGlyoxal (MGO) on rat hepatocytes BRL-3A cells, the viability of the cells was detected by MTT method with 50 渭 mol/L resveratrol treatment BRL-3A, in the presence of MGO, and the concentration of Schiff base in the cells was detected by DNPH method. DCFH-DA fluorescence probe method was used to detect the level of ROS and the content of MDA in lipid peroxidation product. The results showed that MGO significantly increased Schiff base,ROS and MDA in cells and induced oxidative stress in cells. Resveratrol treatment could effectively improve the damage of MGO to cells.
