羰基鎳急性中毒大鼠血清鎳變化與淋巴細(xì)胞DNA損傷及加合物形成的研究
[Abstract]:Objective to observe the changes of serum nickel and lymphocyte DNA damage and the formation of adducts in rats with acute carbonyl nickel poisoning, and to explore the mechanism of lymphocyte DNA damage induced by acute carbonyl nickel poisoning in rats. Methods on the 3rd and 7th day after exposure, serum nickel was measured in 4 groups (low, middle and high concentrations of carbonyl nickel and chlorine in group D) and normal control group (group E). The DNA adducts of lymphocytes were determined by UV absorption spectrometry and DNA damage was observed by single cell gel electrophoresis. Results Serum nickel was highest on the first day after exposure, and decreased gradually with time. The highest absorption peak of lymphocyte DNA was at 259 nm in each exposed group, and the shift of the maximum absorbance of lymphocyte DNA in 1 nm.A group was lower than that in E group (260 nm) on the 3rd day, and the maximum absorbance of Lymphocyte DNA decreased to the lowest point on the 3rd day, compared with that of E group (260 nm). Group B reached the lowest point on the 7th day, group C and group D reached the lowest point on the second day, compared with group E, the difference was statistically significant (P0.05). The tail length of DNA comet and the tail moment of Olive in lymphocytes of each exposed group were higher than those of normal control group (P0.05). Conclusion the serum nickel concentration was the highest in rats on day 1 after acute exposure to carbonyl nickel. The lymphocyte DNA adducts were most formed on the second day after exposure, and the DNA damage on the third day was the most serious, and recovered on the 7th day.
