α-硫辛酸對糖尿病大鼠視網(wǎng)膜VEGF表達(dá)的影響及機制

【摘要】:目的研究α-硫辛酸(α-LA)對糖尿病大鼠視網(wǎng)膜病變及血管內(nèi)皮生長因子(VEGF)表達(dá)的影響及機制。方法選擇72只健康的雄性Wistar大鼠作為實驗材料,將72只大鼠分為4組:空白對照組(A組)12只,糖尿病模型組(B組)24只,α-LA治療組(C組)24只,葡萄糖干預(yù)組(D組)12只,B~D組大鼠按照60mg/kg的劑量腹腔注射鏈脲佐菌素(STZ),C組用100mg/kg的α-LA灌胃,D組用5.0%的葡萄糖溶液灌胃。比較各組大鼠的體質(zhì)量、空腹血糖(FPG)、空腹胰島素(FINS),穩(wěn)態(tài)胰島素抵抗指數(shù)(HOMA-IR)、VEGF表達(dá)、超氧化鈉歧化酶(SOD)、谷胱甘肽(GSH)和IL-6水平,比較B~D組大鼠的GR分期。結(jié)果72h后,B~D組大鼠模型構(gòu)建成功,A組大鼠FPG為(4.57±0.15)mmol/L,B組為(21.72±4.28)mmol/L,C組為(21.54±4.96)mmol/L,D組為(21.83±4.77)mmol/L,A組與B、C組比較差異有統(tǒng)計學(xué)意義(P0.05);A組大鼠體質(zhì)量(210.5±5.2)g,B組(211.2±5.7)g,C組(209.8±5.8)g,D組(208.7±3.4)g,4組的差異無統(tǒng)計學(xué)意義(P0.05)。4組大鼠4、8、12周的體質(zhì)量、FPG、FINS、HOMA-IR、VEGF表達(dá)、SOD、GSH和IL-6水平差異有統(tǒng)計學(xué)意義(P0.05)。12周后,B組、C組和D組大鼠的眼底鏡檢查GR分期差異有統(tǒng)計學(xué)差異(P0.05)。結(jié)論α-LA的應(yīng)用可以有效抑制大鼠視網(wǎng)膜VEGF的表達(dá),這一作用可能與改善胰島素抵抗、抑制慢性炎癥和氧化應(yīng)激多個環(huán)節(jié)相關(guān)。
[Abstract]:Objective to study the effect and mechanism of 偽-lipoic acid (偽-LA) on the expression of vascular endothelial growth factor (VEGF) and retinopathy in diabetic rats. Methods Seventy-two healthy male Wistar rats were divided into 4 groups: control group (group A, n = 12), diabetic model group (group B, n = 24) and 偽 LA treatment group (group C, n = 24). There were 12 rats in glucose intervention group (group D). Rats in group D received intraperitoneal injection of streptozotocin (STZ), C) with 偽-LA of 100mg/kg according to the dose of 60mg/kg. Group D was given 5. 0% glucose solution. The body weight, fasting insulin (FINS), steady-state insulin resistance index (HOMA-IR), VEGF) expression, superoxide dismutase (SOD), glutathione (GSH) and IL-6 levels were compared in each group, and GR stages of BSD group were compared. Results after 72 hours, the FPG of group A was (4.57 鹵0.15) mmol/L,B, (21.72 鹵4.28) mmol/L,C, (21.54 鹵4.96) mmol/L,D was (21.83 鹵4.77) mmol/L,A and group C was (21.83 鹵4.77) mmol/L,A. There was no significant difference between group B and group B (P 0.05, 210.5 鹵5.2g / g, P 0.05, 211.2 鹵5.7g / g, P 0.05). There was no significant difference between group A and group B (209.8 鹵5.8g / g, P 0.05). After 12 weeks, there were significant differences in body mass, FPG,FINS,HOMA-IR,VEGF expression, SOD,GSH and IL-6 levels in rats (P0.05). After 12 weeks, there were significant differences in GR staging between group B, group C and group D (P0.05). Conclusion 偽-LA can effectively inhibit the expression of VEGF in rat retina, which may be related to improving insulin resistance, inhibiting chronic inflammation and oxidative stress.
