維生素D及其受體對(duì)RIP1調(diào)控作用的研究

【摘要】:目的探討維生素D及其受體對(duì)受體相互作用蛋白(RIP)1表達(dá)的影響及其作用機(jī)制。方法采用實(shí)時(shí)定量PCR和Western blot技術(shù),檢測(cè)維生素D刺激對(duì)RIP1的m RNA和蛋白表達(dá)水平的影響,采用免疫共沉淀的方法探討維生素D及其受體對(duì)RIP1的調(diào)節(jié)機(jī)制。結(jié)果維生素D及其受體能夠顯著降低RIP1的m RNA及蛋白水平表達(dá),抑制HSP90與RIP1復(fù)合物的形成。結(jié)論維生素D及其受體通過(guò)調(diào)節(jié)HSP90與RIP1的相互作用調(diào)控RIP1的表達(dá),為維生素D及其受體抑制腫瘤的發(fā)生提供了新的理論依據(jù)。
[Abstract]:Objective to investigate the effect of vitamin D and its receptor on the expression of receptor interacting protein (RIP) 1 and its mechanism. Methods the effects of vitamin D stimulation on the expression of m RNA and protein in RIP1 were detected by real-time quantitative PCR and Western blot. The regulation mechanism of vitamin D and its receptors on RIP1 was investigated by immunoprecipitation. Results Vitamin D and its receptor could significantly reduce the expression of m RNA and protein in RIP1 and inhibit the formation of HSP90 / RIP1 complex. Conclusion Vitamin D and its receptors regulate the expression of RIP1 by regulating the interaction between HSP90 and RIP1, which provides a new theoretical basis for the inhibition of tumor development by vitamin D and its receptors.
