離心造粒法制備奧美拉唑速釋微丸

【摘要】:采用離心造粒法制備奧美拉唑(1)速釋微丸,并用單因素法優(yōu)化處方和工藝參數(shù)。處方中抗酸劑(碳酸氫鈉)用于提高局部微環(huán)境的p H值,以提高1在酸性條件下的穩(wěn)定性;此外可起到與崩解劑羧甲基淀粉鈉相似的加快藥物溶出的作用。優(yōu)化處方宜采用28目的微晶纖維素空白丸芯為母核,以1.5%羥丙甲纖維素為黏合劑,碳酸氫鈉和微晶纖維素PH101(填充劑)用量為200和60 g。優(yōu)化后,18~24目1速釋微丸的收率為(89.6±2.5)%,在pH 1.2鹽酸中15 min時1的溶出率達(dá)85%以上,且1 h內(nèi)穩(wěn)定性良好。
[Abstract]:Omeprazole (1) pellets were prepared by centrifugal granulation, and the formulation and technological parameters were optimized by single factor method. The antiacid agent (sodium bicarbonate) was used to improve the pH value of local microenvironment in order to improve the stability of 1 in acidic condition and to accelerate the dissolution of the drug similar to the disintegrating agent sodium carboxymethyl starch. The optimum formulation is to use 28 target microcrystalline cellulose blank pellets as mother nucleus, 1.5% hydroxypropyl methyl cellulose as binder, and the dosage of sodium bicarbonate and microcrystalline cellulose PH101 as 200 and 60 g. The yield was (89.6 鹵2.5)%, and the dissolution rate of 1 in pH 1.2 hydrochloric acid for 15 min was more than 85%, and the stability was good within 1 h.
