【摘要】：目的設(shè)計(jì)合成新型代謝綜合征治療藥物。方法以具有降脂活性的阿昔莫司做為基本結(jié)構(gòu)單元,與貝特類藥物的"苯氧脂酸類"藥效團(tuán)有機(jī)結(jié)合,設(shè)計(jì)了一類新型的苯氧脂肪酸酯類化合物,以對(duì)羥基苯胺、對(duì)羥基芐胺以及對(duì)羥基苯乙胺為原料,首先將氨基保護(hù),然后與相應(yīng)的鹵代脂肪鏈羧酸酯成醚,然后脫除Boc保護(hù)基得到相應(yīng)的胺類中間體,最后與阿昔莫司成酰胺得目標(biāo)化合物,并考察了目標(biāo)化合物對(duì)Triton WR-1339所致急性高脂血癥小鼠血清甘油三酯和總膽固醇的影響。結(jié)果和結(jié)論合成目標(biāo)化合物5個(gè),均經(jīng)~1H NMR和ESI-MS確證結(jié)構(gòu)正確。初步活性評(píng)價(jià)結(jié)果表明,所合成化合物均具有一定的降脂活性,其中化合物4c的調(diào)脂活性與陽(yáng)性對(duì)照苯扎貝特接近,對(duì)于新型代謝綜合征藥物的發(fā)現(xiàn)具有積極意義。
[Abstract]:Objective to design a new synthetic drug for metabolic syndrome. Methods A new class of phenoxy fatty acid esters was designed by using acymomus with lipid-lowering activity as the basic structure unit, and combined with the phenoxyllipoxylic acid group of the Bette drugs, and then the phenoxy fatty acid esters were used as p-hydroxyaniline. P-hydroxybenzylamine and p-hydroxyphenylethylamine were used as raw materials to protect the amino group, then to form an ether with the corresponding halogenated fatty chain carboxylic acid ester, and then to remove the Boc protection group to obtain the corresponding amine intermediate. The effects of the target compounds on serum triglyceride and total cholesterol in acute hyperlipidemic mice induced by Triton WR-1339 were investigated. Results and conclusion the structures of the five target compounds were confirmed by 1H NMR and ESI-MS. The results of preliminary activity evaluation showed that all the synthesized compounds had certain lipid-lowering activities, and the lipid-regulating activity of compound 4c was close to that of the positive control, which was of positive significance to the discovery of new drugs for metabolic syndrome.
