氫溴酸東莨菪堿溫敏型鼻用原位凝膠的制備

【摘要】:目的制備氫溴酸東莨菪堿的溫敏型鼻用原位凝膠。方法用不同質(zhì)量濃度和配比的殼聚糖、β-甘油磷酸鈉制備不同pH的原位凝膠,并測定其凝膠化時間、黏度變化情況與載藥凝膠的累積釋放度。結(jié)果質(zhì)量濃度為2%的殼聚糖和56%的β-甘油磷酸鈉體積比為6∶1,pH 7.1時原位凝膠黏度達(dá)到5 Pa·s,初始凝膠溫度為34℃,隨著溫度的升高、殼聚糖質(zhì)量濃度的增加、β-甘油磷酸鈉含量的增加、pH升高(由6.5升至7.2),凝膠化時間減少;以氫溴酸東莨菪堿為模型藥物,載藥凝膠中藥物10h累積釋放度為71%,釋放曲線符合一級動力學(xué)方程。結(jié)論殼聚糖/β-甘油磷酸鈉制備的氫溴酸東莨菪堿溫敏型原位凝膠對藥物具有緩釋作用,適合制成鼻用制劑。
[Abstract]:Objective to prepare a temperature sensitive in situ gel for scopolamine hydrobromide. Methods different in situ gels of pH were prepared by using chitosan and 尾 -glycerophosphate with different concentration and ratio.
