反相高效液相色譜法測(cè)定人體血漿中維拉佐酮的含量

【摘要】:目的:建立反相高效液相色譜法測(cè)定人體血漿中維拉佐酮的含量。方法:色譜柱為美國(guó)Diamonsil~汶(250 mm×4.6mm,5μm)C_(18)反相柱,流動(dòng)相為乙腈-甲醇-緩沖液(30∶30∶40,V/V/V),流速為1.0mL·min~(-1),柱溫30℃;檢測(cè)波長(zhǎng)240 nm。結(jié)果:維拉佐酮血漿濃度在0.16~32.00mg·L~(-1)范圍內(nèi),線(xiàn)性關(guān)系良好(r=0.998 6,n=8),平均日間精密度為4.10%,平均相對(duì)回收率為97.85%。結(jié)論:本方法準(zhǔn)確、快速、簡(jiǎn)便,適于人血漿中維拉佐酮的血藥濃度檢測(cè)和藥動(dòng)學(xué)研究。
