【摘要】：目的建立1種同時(shí)測定骨瓜提取物注射劑中15種氨基酸含量的RP-HPLC法。方法以異硫氰酸苯酯(PITC)為柱前衍生試劑對樣品中15種氨基酸進(jìn)行衍生化,采用Ocean C18(250 mm×4.6 mm,5μm)色譜柱,流動(dòng)相A為水-乙腈(體積比1∶4),流動(dòng)相B為濃度0.1 mol·L-1乙酸鈉-乙腈(體積比97∶3),梯度洗脫,流速為1.0 m L·min-1,柱溫為25℃,檢測波長為254 nm,進(jìn)樣量為10μL。用內(nèi)標(biāo)法計(jì)算15種氨基酸的含量。結(jié)果 15種氨基酸質(zhì)量濃度在1.24~160.72 mg·L-1內(nèi)與峰面積呈良好的線性關(guān)系(r=0.997 7~1.000 0),15種氨基酸的平均加樣回收率在97.7~102.4%內(nèi),加樣回收率的RSD值在1.7%~3.3%內(nèi)。結(jié)論該方法準(zhǔn)確、可靠、靈敏,可以用于同時(shí)測定骨瓜提取物注射劑中15種氨基酸的含量。
[Abstract]:Objective to establish a RP-HPLC method for simultaneous determination of 15 amino acids in extract injection of Gugu melon. Methods 15 kinds of amino acids in the sample were derivatized with phenylethyl isothiocyanate (PITC) as precolumn derivatization reagent.
