干擾素誘導(dǎo)的跨膜蛋白抗病毒作用的研究進(jìn)展

【摘要】:干擾素誘導(dǎo)的跨膜蛋白(Interferon-induced Transmembrane Proteins,IFITMs)是20世紀(jì)80年代發(fā)現(xiàn)的一種宿主限制因子蛋白,1996年發(fā)現(xiàn)該蛋白具有抗病毒作用,目前該蛋白的抗病毒作用及其作用機制已成為研究熱點。研究表明IFITM能抑制多種病毒的復(fù)制,包括甲型流感病毒、人類免疫缺陷病毒-1、丙型肝炎病毒、埃博拉病毒和西尼羅病毒等。IFITM蛋白主要在病毒生命周期的早期,即病毒進(jìn)入細(xì)胞質(zhì)之前,發(fā)揮抑制病毒復(fù)制的作用。近來的研究表明,IFITM蛋白通過影響病毒包膜與內(nèi)涵體膜的融合抑制病毒復(fù)制,但具體機制尚不明確。本文對IFITM的發(fā)現(xiàn)、結(jié)構(gòu)、抗病毒作用以及潛在的作用機制進(jìn)行了綜述。
[Abstract]:Interferon-induced transmembrane protein (Interferon-induced Transmembrane Proteins,IFITMs) is a host limiting factor protein discovered in 1980s. It was found in 1996 that the protein has antiviral effect. At present, the antiviral effect and its mechanism of IFN- induced transmembrane protein have become a hot research topic. Studies have shown that IFITM inhibits the replication of a variety of viruses, including influenza A, human immunodeficiency virus -1, hepatitis C virus, Ebola virus and West Nile virus proteins, mainly in the early stages of the virus's life cycle. That is, the virus before entering the cytoplasm, play a role in inhibiting virus replication. Recent studies have shown that the IFITM protein inhibits viral replication by affecting the fusion of viral envelope and connotative membrane, but the specific mechanism remains unclear. The discovery, structure, antiviral effect and potential mechanism of IFITM are reviewed in this paper.
